摘  要:煤炭开采造成大面积矿区土地塌陷,导致当地生态系统的脆弱性显著增加。煤矸石充填复垦是修复矿区塌陷地的常用方法,土壤细菌群落对于反映复垦土壤质量和生态健康状况具有重要的指示作用。为了了解矿区塌陷地煤矸石充填复垦对土壤细菌群落的影响,采用野外采样和高通量lllumina Miseq 16S rRNA测序技术,以周边普通耕地作为对照组(CK),分析了中国东部典型高潜水位矿区采煤塌陷地煤矸石充填复垦耕地(G)不同土层深度(0~20,20~40,40~60 cm)下土壤理化性质、土壤细菌群落多样性和结构组成的变化,以及土壤细菌群落与土壤理化性质的关系。结果表明:①煤矸石充填复垦显著改变了研究区土壤理化性质,不同土层深度下土壤性质变化趋势差异明显;②煤矸石充填复垦地的土壤细菌群落多样性与丰富度均低于普通耕地,且与土层深度呈显著负相关(P<0.05),40~60 cm层因煤矸石充填造成细菌群落组成差异大于其他土层。另外,变形菌门、放线菌门和酸杆菌门是所有处理中的优势菌门;③典型相关分析(CCA)和相关性分析(Pearson)结果表明,pH、含水量(MoiC)、有机碳(SOC)、速效磷(AP)和速效钾(AK)是影响土壤细菌群落组成的主要土壤理化性质,优势菌门仅与pH,AP,MoiC等部分理化性质显著相关。Coal mining often causes large areas of subsidence land collapse in mining areas,leading to a significant increase in the vulnerability of the local ecosystem.Reclamation with coal gangue filling is commonly adopted to repair the subsidence of mining areas,and soil bacterial community plays an important indicative role in reflecting the quality and ecological health of reclaimed soil.In order to understand the impact of reclamation of subsidence area with coal gangue filling on soil bacterial community,takeing surrounding normal cultivated land(CK)as controla comparison,field sampling and high throughput Illumina Miseq 16S rRNA gene sequencing were used to analyze the soil physical and chemical properties,the variations of soil bacterial community composition and diversity,and the relationship between them in different soil depths(0-20,20-40 and 40-60 cm)of reclaimed land filled with coal gangue(G).The results showed that:①Ccoal gangue reclamation significantly changed the soil physical and chemical properties in the study area,the changing trend of soil properties under different soil depths was obviously different.②The diversity and abundance of soil bacterial community in the reclaimed land filled with coal gangue were lower than those in normal cultivated lands,which also showed a significant negative correlation with soil depth(P<0.05).The composition difference of bacterial community in the 40-60 cm soil layer due to coal gangue filling was greater than that in other soil layers.Besides,Proteobacteria,Actinobacteria,and Acidobacteria were the dominant phyla in all treatments.③Results from canonical correlation analysis(CCA)and Pearson correlation analysis indicated that pH,Moisture Content(MoiC),Soil Organic Carbon(SOC),Available Phosphorus(AP),and Available Potassium(AK)were the main soil physical and chemical properties in shaping soil bacterial communities,and the dominant bacteria phyla were only significantly associated with some soil properties such as pH,AP and MoiC.
