摘  要:目的:采用超高效液相色谱-四级杆-静电轨道串联高分辨质谱法(UPLC-QE-Orbitrap-MS)对和血止痒方的化学成分进行表征。方法:采用沃特世ACQUITY UPLC@Hss T3型色谱柱(100 mm×2.1 mm,1.7μm),以乙腈-0.1%甲酸为流动相梯度洗脱,流速为0.2 mL·min^(-1);采用加热电喷雾离子源(HESI),正、负离子模式分别进行一级、二级质谱数据采集;采用数据库和文献报道对和血止痒方的化学成分进行表征。结果:在和血止痒方中共鉴定出92个化合物,结合数据库和文献报道对其中67个化合物进行初步表征,包括萜类、苯丙素类、有机酸类、苯酞类、生物碱类、黄酮类及其他类,并对化合物的药材来源进行归属。结论:该方法灵敏度高,分析快速,结果准确、可靠,能够全面快速地分析和鉴定和血止痒方中的化学成分,为其进一步的药效物质基础及质量控制研究提供了参考。Objective:To characterize the chemical constituents of Hexue Zhiyang Formula(HZF)by ultra-high performance liquid chromatography-quadrupole-orbitrap mass spectrometry(UPLC-QE-Orbitrap-MS).Methods:Components underwent chromatographic separation with Waters ACQUITY UPLC@Hss T3 column(100 mm×2.1 mm,1.7μm).The mobile phase consisted of acetonitrile-water(including 0.1%formic acid)for gradient elution,and the flow rate was 0.2 mL·min^(-1).MS and MS/MS data were collected in the positive and negative ion modes using heated electrospray ionization(HESI)source.The chemical constituents of HZF were characterized based on databases and literature reports.Results:Ninety-two compounds were identified in HZF,and 67 compounds were primarily characterized according to databases and literature reports,including terpenoids,phenylpropanoids,organic acids,phthalides,alkaloids,and flavonoids.Additionally,the original medicinal materials of compounds were assigned.Conclusion:This method is high in sensitivity,fast in analysis,and accurate and reliable in results,and can comprehensively and quickly analyze and identify the chemical components in HZF,which is expected to provide references for further investigation on the material basis and quality control of HZF.
