摘 要:目的:建立超高效液相色谱法(UPLC)同时测定不同产地野菊花中绿原酸、木犀草苷、木犀草素、蒙花苷和芹菜素5个成分的含量,为野菊花的质量控制和评价提供依据。方法:采用ACQUITY UPLC CSH C18(150 mm×2.1 mm,1.7μm)色谱柱,柱温为(30±5)℃,进样体积为2μL,检测波长为326 nm,以乙腈-0.02%磷酸溶液为流动相梯度洗脱。结果:5个成分在相应的质量浓度范围内与峰面积的线性关系均良好(r≥0.9990),精密度、稳定性、重复性试验RSD均小于2.00%,39批次野菊花中绿原酸、木犀草苷、蒙花苷、木犀草素、芹菜素质量分数分别为0.30~5.40、0.10~2.80、0~15.40、0.60~11.90、0~1.10 mg·g^(-1)。结论:此方法方便、准确、稳定,可用于同时测定野菊花中5个成分的含量。Objective:To establish an ultra-high performance liquid chromatography(UPLC)method for simultaneous determination of chlorogenic acid,luteoloside,luteolin,buddleoside,and apigenin in Chrysanthemum indicum from different habitats to provide references for quality control and evaluation of C.indicum.Methods:Components underwent chromatographic separation with the ACQUITY UPLC CSH C18 column(150 mm×2.1 mm,1.7μm)at(30±5)℃.The mobile phase was composed of acetonitrile and 0.02%phosphate solution for gradient elution.The detection wavelength was 326 nm and the injection volume was 2μL.Results:The five components showed good linearity with peak areas at the corresponding mass concentrations(r≥0.9990).The RSD values of precision,stability,and repeatability were all less than 2.00%.The mass fractions of chlorogenic acid,luteoloside,buddleoside,luteolin,and apigenin in 39 batches of samples were 0.30-5.40,0.10-2.80,0-15.40,0.60-11.90,and 0-1.10 mg·g^(-1),respectively.Conclusion:This method is rapid,accurate,and stable,and can be used to simultaneously determine the content of five components in C.indicum.
