摘 要:运用生物信息学方法对2型糖尿病(T2DM)易感基因进行筛选、分析,从分子水平探讨T2DM发病机制。利用GEO数据库查找T2DM患者外周血单个核细胞差异表达的基因集;利用DAVID数据库进行GO功能富集分析、KEGG信号通路分析;利用STRING工具与Cytoscape软件及MCODE插件对差异表达基因做蛋白质互作网络分析。结果如下:1)GO功能富集分析显示T2DM差异表达基因主要分布在G蛋白偶联受体信号通路、细胞增殖分化、信号转导等生物过程中,大部分与神经传导相关;2)KEGG信号分析结果显示差异表达基因主要集中在神经活性配体-受体相互作用信号通路、Ras信号通路等通路;3)共得到可信度较高的RAC1、PIK3R1、CDC42等12个关键基因。可见T2DM的发病受到炎症反应、氧化应激及神经信号转导等多种因素的影响。Based on the screening and analysis of the susceptibility genes of Type 2 Diabetes Mellitus(T2DM),this paper explores the pathogenesis of T2DM by using bioinformatics analysis method.Within the GEO database,we find the gene set of differential expression of peripheral blood mononuclear cells in type 2 diabetic patients.DAVID database is used for the analysis of GO function enrichment and KEGG signal pathway.String,Cytoscape and MCODE plug-in are used to analyze protein interaction network of differentially expressed genes.The results are as follows:1)GO functional enrichment analysis shows that those differentially expressed genes play a major role in biological processes such as G protein-coupled receptor signaling pathway,cell proliferation and differentiation,signal transduction,most of which are related to neuritis conduction;2)The results of KEGG signal analysis show that differentially expressed genes are mainly distributed in the neuroactive ligand-receptor interaction signaling pathway and Ras signaling pathway;3)12 key genes including RAC1,PIK3R1,CDC42,etc.are reliably obtained.Hence,we come to the conclusion that the pathogenesis of T2DM involves many factors,such as inflammatory response,oxidative stress,hormone stimulation response change,and neural signal transduction,etc.
