摘  要:为探究陕西省安康市林麝死亡原因,本试验无菌采集病死林麝肺脏进行细菌的分离、鉴定及耐药性与致病性分析。通过病理解剖、细菌分离纯化、革兰染色镜检、生化鉴定及16S rRNA基因测序等方法对分离株进行鉴定;通过药敏纸片法和毒力基因PCR扩增分析分离株的耐药性和致病性,并对其进行分子分群。结果显示,在死亡林麝肺脏分离出1株革兰阴性短杆菌,经鉴定为大肠埃希菌,命名为LS202010;进化树分析结果显示,分离株LS1与挪威分离株LR883965.1同源性最高;药敏试验结果显示,分离株对阿莫西林、复方阿莫西林、泰万菌素、氨苄西林、泰乐菌素、氨苄青霉素、头孢噻呋钠、金霉素、新霉素、复方磺胺氯达嗪钠、安普霉素等耐药;毒力基因PCR分析结果显示,该菌含有etrA毒力基因,表明该菌属于肠聚集性大肠埃希菌;分子分群结果显示,该菌属于B1群。本试验分离出1株林麝源大肠埃希菌LS202010,并证明其具有较高的致病性和耐药性,为致病性大肠埃希菌的防制奠定理论基础。To explore the cause of death of forest musk deer in Ankang City of Shaanxi Province in China,the lungs of dead forest musk deer were collected aseptically for bacterial isolation.The isolated bacteria were analyzed and identified through anatomical observation,bacterial isolation and purification,Gram staining and microscopic examination,biochemical identification and 16 S rDNA sequencing.Drug resistance and pathogenicity of the isolates was analyzed by disk diffusion test and PCR assay of virulence gene,and phylo-typing analysis.The results showed that a Gram-negative Brevibacterium was isolated from the dead forest musk deer lungs and was identified as Escherichia coli(E.coli)and named as LS202010.The phylogenetic tree analysis showed that the LS202010 had the highest homology with LR883965.1 isolate from Norway.The result of drug sensitive test showed that the strain was resistant to amoxicillin,compound amoxicillin,tylvalosin,ampicillin,tylosin,ampicillin,ceftiofur sodium,aureomycin,neomycin,compound sulfachloropyridazine sodium and apramycin.The PCR assay of virulence gene showed that the isolate LS202010 contains the virulence gene etrA.The result of phylo-typing assay showed that the strain belonged to the B1 group.In conclusion,a strain of enteroaggregative E.coli was isolated from forest musk deer,and it was proved to have high pathogenicity and multiple drug resistance,which laid a foundation for the prevention and treatment of pathogenic E.coli.
