3例犬皮肤肥大细胞瘤病理学及c-kit免疫组织化学染色研究

摘　　要：为了对临床送检的3例犬皮肤肿物进行确诊与预后评估,本研究对送检样品制作组织切片,HE染色后在显微镜下可见大量增生的细胞,呈现"煎蛋样"形态,胞浆内含有蓝灰色颗粒物质,符合肥大细胞的特征;进而采用甲苯胺蓝染色,观察到肿瘤细胞胞浆内颗粒物质呈蓝紫色的特异性着色,进一步证实其为肥大细胞起源;最后通过c-kit免疫组织化学染色判断免疫表型,可见肿瘤细胞KIT蛋白呈不同标记类型的阳性着色。通过病理学分析及c-kit染色,确定3个病例均为皮肤型肥大细胞瘤(cutaneous mast cell tumor,cMCT)。研究结果不仅可对临床病例的治疗方案设计与预后评估提供参考,而且可为犬肥大细胞瘤的确诊提供可靠的技术方法。To diagnose and evaluate the prognosis of three cases of canine skin neoplasm,HE staining of the tissue sections were performed.Under microscope,proliferation of tumor cells characterized as"fried egg like"shape and cytoplasm contained blue gray particulate matter,which was consistent with the characteristics of mast cells.Next,toluidine blue staining was used,specific blue-purple staining of granular material within the cytoplasm of the tumor cells was observed,further confirming their mast cell origin.Finally,c-kit immunohistochemical staining was performed to determine the immunophenotype,and it was found that the KIT protein of tumor cells showed different types of positive staining,which could provide a basis for prognostic evaluation.Through pathological observation and c-kit immunohistochemical analysis,all three cases were diagnosed as cutaneous mast cell tumor(cMCT).The results of this study can not only provide reference for the treatment plan design and prognosis evaluation of these clinical cases,but also provide a reliable technical method for the diagnosis of cMCT.
