摘 要:哺乳动物毛纤维在人类生活中发挥着不可或缺的作用,已发现300多个基因与哺乳动物皮肤、眼睛和毛发颜色相关,而研究证实miRNA对基因有着重要的调控功能。本试验前期研究发现miR-370在白色和棕色绒山羊皮肤组织中差异表达,但其如何通过调控基因表达影响毛色仍未知。本试验通过生物信息学软件筛选出miR-370的靶基因Wnt3a和KITLG,利用qPCR和免疫组化技术分析Wnt3a和KITLG在白色、棕色绒山羊皮肤中的表达、分布情况及相互关系,同时测定不同毛色皮肤黑色素的含量。结果显示,棕色绒山羊皮肤黑色素含量显著高于白色绒山羊皮肤黑色素含量;Wnt3a和KITLG主要表达于绒山羊皮肤的基底膜和内、外根鞘,且在棕色绒山羊皮肤的表达显著高于白色绒山羊,与miR-370的表达趋势相同,表明miR-370通过调控Wnt3a和KITLG参与绒山羊毛色的形成。Mammalian hair fibers play an indispensable role in human life.More than 300 genes have been found to be related to skin,eye and hair color of mammals,and studies have confirmed that miRNAs play an important regulatory role in genes.Previous studies found that miR-370 was differentially expressed in the skin tissues of white and brown Cashmere goats,but how miR-370 affects hair color through regulating gene expression is still unknown.The target genes Wnt3 a and KITLG were predicted by bioinformatics software.The expression,distribution and correlation of Wnt3 a and KITLG in white and brown Cashmere goat skin were analyzed by qPCR and immuno-histochemical techniques,and the content of melanin in different skin color was determined.The results showed that the melanin content in brown Cashmere goat skin was significantly higher than that in white Cashmere goat skin.Wnt3 a and KITLG were mainly expressed in the basement membrane and inner and outer root sheath of Cashmere goat skin,and the expression of Wnt3 a and KITLG in brown Cashmere goat skin was significantly higher than that in white Cashmere goat skin,which was similar to the expression trend of miR-370.These results suggested that miR-370 was involved in the formation of wool color of Cashmere goats by regulating Wnt3 a and KITLG.
