摘 要:目的研究PEI-SPIO标记的人羊膜间充质干细胞(human amnion mesenchymal stem cells,hAMSCs)移植对小鼠创伤愈合的影响。方法酶消化法分离hAMSCs后PEI-SPIO标记,普鲁士蓝染色检测hAMSCs的标记率,WST-1检测标记细胞增殖能力,透射电镜观察标记细胞内铁离子的分布,建立小鼠全皮层创伤模型,皮内移植PEI-SPIO标记的hAMSCs,观察创面愈合及移植的hAMSCs的迁移情况。结果 PEI-SPIO标记对hAMSCs细胞形态无明显改变,标记率99%,PEI-SPIO标记浓度小于10μg/ml时,对hAMSCs的增殖无影响。体内实验证明PEI-SPIO标记的hAMSCs皮内移植能促进小鼠皮肤创伤愈合,普鲁士蓝染色可示踪移植的PEI-SPIO标记的hAMSCs。结论 PEI-SPIO可以作为hAMSCs治疗创伤愈合的标记物。Objective This study aimed to investigate the effect of transplantion of PEI-SPIO labeled hAMSCs on wound healing of mice. Methods hAMSCs were isolated by enzymatic digestion, labeled by PEI-SPIO and observed by inverted phase contrast microscope. Then we stained labeled hAMSCs by Prussian blue, detected the proliferation ability of the labeled cells by WST-1 assay, observed the distribution of iron ions in the labeled cells by transmission electron microscopy. Furthermore we transplanted PEI-SPIO labeled hAMSCs in a mouse model of cutaneous wounds to observe wound healing and the migration of hAMSCs. Results The morphology of hAMSCs labeled by PEI-SPIO was not significantly changed, and the labeling rate was 99%. The proliferation of hAMSCs was not affected when the concentration of PEI-SPIO was less than 10μg/ml. In vivo experiments demenstrated that PEI-SPIO-labeled hAMSCs could promote skin wound healing in mice after intradermal transplantation, and Prussian blue staining could trace the PEI-SPIO-labeled hAMSCs. Conclusion PEI-SPIO has the potential to be used for labeling hAMSCs in the treatment of wound healing.
