摘 要:目的探讨疏风解毒胶囊对慢性阻塞性肺病(COPD)小鼠肺部氧化损伤组织修复作用及免疫功能影响。方法 50只小鼠随机分为空白组、模型组和疏风解毒胶囊0.1、0.2、0.5 g/kg组,经烟熏法联合脂多糖建立COPD模型。以Buxco系统检测小鼠肺功能指标(PIF、MV、PEF)后处死小鼠,取肺组织切片行HE染色观察病理学变化,采用酶联免疫吸附法检测血清氧化指标(SOD、MDA、GSH、TNF-α、hs-CRP),采用流式细胞仪检测血清免疫功能指标(CD3^(+)、CD4^(+)、CD8^(+)、CD4^(+)/CD8^(+))。结果与空白组比较,模型组肺组织出现明显病理损伤及坏死,肺功能指标PIF、MV、PEF及CD3^(+)、CD4^(+)、CD4^(+)/CD8^(+)水平及SOD、GSH-Px活性明显降低,CD8^(+)、TNF-α、hs-CRP、MDA明显升高(P<0.05)。与模型组相比,疏风解毒胶囊0.2、0.5 g/kg组肺组织病理损伤明显减轻;且肺功能指标PIF、MV、PEF及CD3^(+)、CD4^(+)、CD4^(+)/CD8^(+)水平及SOD、GSH-Px活性明显升高,CD8^(+)、TNF-α、hs-CRP、MDA明显降低(P<0.05)。结论疏风解毒胶囊能有效调节COPD小鼠的氧化应激损伤及免疫功能,可减轻肺组织病理形态损伤。。Objective To investigate the effect of Shufeng Jiedu Capsule on lung oxidative damage tissue repair and immune function in mice with chronic obstructive pulmonary disease(COPD). Methods 50 mice were randomly divided into blank group, model group and Shufeng Jiedu capsule 0.1, 0.2 and 0.5 g/kg groups, COPD model was established by fumigation combined with lipopolysaccharide. The lung function indexes(PIF, MV and PEF) of mice were detected by Buxco system, then the mice were sacrificed. The pathological changes of lung tissue were observed by HE staining, which the serum oxidation indices(SOD, MDA, GSH, TNF-α and hs-CRP) were detected by enzym-linked immunosorbent assay, and the serum immune function indices(CD3^(+), CD4 ^(+), CD8 ^(+), CD4 ^(+)/CD8^(+)) were detected by flow cytometry. Results Compared with blank group, lung tissue in model group showed obvious pathological damage and necrosis, and lung function indexes(PIF, MV, PEF, Cd3 ^(+), Cd4 ^(+), Cd4 ^(+)/CD8^(+)) and activities of SOD and GSH-Px were significantly decreased, while CD8^(+), TNF-α, hs-CRP and MDA were significantly increased(P<0.05). Compared with the model group, the pathological injury of lung tissue in 0.2 and 0.5 g/kg Shufengjiedu capsule groups was remarkably reduced, and lung function indexes PIF, MV, PEF, Cd3 ^(+), Cd4^(+), Cd4 ^(+)/CD8^(+) levels and activities of SOD and GSH-Px were significantly increased, while CD8 ^(+), TNF-α, hs-CRP and MDA were significantly decreased(P<0.05). Conclusion Shufengjiedu capsule can effectively regulate oxidative stress injury and immune function of COPD mice, which it can also reduce pathological pathological damage of lung tissue.
