摘  要:目的探讨过表达Cdh1蛋白对高糖诱导的视网膜Müller细胞株GFAP表达的影响。方法培养的Müller细胞分为4组:对照组(Control)、高糖组(HG group,30 mmol/L葡萄糖)、Cdh1过表达组(Cdh1,30mmol/L葡萄糖+过表达Cdh1蛋白的慢病毒载体)、病毒阴性对照组(NC-Cdh1,30 mmol/L葡萄糖+慢病毒载体稀释液),显微镜下观察Müller细胞一般生长状况,免疫荧光检测GFAP在Müller细胞的表达情况,Western blot检测Müller细胞GFAP及Cdh1蛋白的相对表达。结果 HG组和NC-Cdh1组GFAP表达水平明显高于Control组,Cdh1明显低于Control组(P<0.05);Cdh1组GFAP表达水平明显低于HG组,Cdh1高于HG组(P<0.05),而HG组和NCCdh1组之间无统计学意义(P> 0.05)。结论过表达Cdh1蛋白下调高糖诱导的视网膜Müller细胞GFAP表达,对高糖诱导的Müller细胞损伤起到保护作用。Objective To investigate the effect of overexpressed Cdh1 protein on the expression of GFAP in retinal Müller cell line induced by high glucose. Methods The cultured Müller cells were divided into four groups: control group(Control), high glucose group(HG, 30 mmol/L glucose), Cdh1 overexpression group(Cdh1, 30 mmol/L glucose+lentivirus vector overexpressing Cdh1 protein), virus negative control group(NC-Cdh1, 30 mmol/L glucose+Lentivirus vector diluent). Microscope was used to observe the general growth of Müller cells, the expression of GFAP in Müller cells was detected by immunofluorescence, and the relative expression of GFAP and Cdh1 proteins in Müller cells was detected by Western blot. Results The expression level of GFAP in HG group and NC-Cdh1 group was significantly higher than that in control group, and the expression level of Cdh1 was significantly lower than that in control group(P<0.05);the expression level of GFAP in Cdh1 group was significantly lower than that in HG group, and the expression level of Cdh1 was significantly higher than that in HG group(P<0.05), but with no statistical significance between HG group and NC-Cdh1 group(P>0.05). Conclusion Overexpressed Cdh1 protein down-regulated the expression of GFAP in retinal Müller cells induced by high glucose, and plaid a protective role in the damage of Müller cells induced by high glucose.
