摘 要:目的:探讨厚朴酚对H1N1所致流感病毒性肺炎的防治作用及其作用机制。方法:60只小鼠随机分成正常对照组、模型组、厚朴酚低剂量组(96 mg/kg)、中剂量组(192 mg/kg)、高剂量组(384 mg/kg)和阳性(利巴韦林,180 mg/kg)组,每组10只。通过流感病毒鼻腔攻毒建立感染小鼠病毒性肺炎模型,同时灌胃相应药物或纯净水;采用HE染色观察各组小鼠肺组织肺泡的保护作用;采用Western Blot法检测各组小鼠肺组织Toll样受体7(TLR7)、细胞内髓样分化因子88(MyD88)、核因子-κBp65(NF-κBp65)、肿瘤坏死因子-α(TNF-α)蛋白表达的变化。结果:厚朴酚各剂量组均能不同程度减轻肺组织肺泡损伤,降低TLR7、MyD88、NF-κBp65、TNF-α的表达。结论:厚朴酚能抑制流感(H1N1)所致病毒性肺炎的炎症反应。Objective:To investigate the intervention effect of Magnolol on viral pneumonia caused by influenza(H1N1)and its mechanism.Methods:60 mice were divided into normal,model,low,middle,high dose Magnolol(96,192,384 mg/kg)and positive(Ribavirin,180 mg/kg)groups with each 10 mice.The model of viral pneumonia was established by nasal challenge of influenza virus,and Magnolol,Ribavirin and purified water was given the corresponding group respectively by gavage.The method of hematoxylin eosin staining was used to investigate the protective effect of each group on pulmonary alveoli.Western blot was used to analyse the protein levels of TLR7 MyD88 NF-κBp65 and TNF-α.Results:Magnolol can reduce the alveolar injury of lung tissue in different degrees,Magnolol with in 96~384 mg/kg could suppress the levels of TLR7 MyD88 NF-κBp65 and TNF-α.Conclusion:Magnolol can inhibit the inflammatory reaction of viral pneumonia caused by influenza(H1N1).
