摘  要:目的探讨p53在动脉钙化中对平滑肌细胞向成骨样细胞转分化的影响。方法21只健康雄性apoE-/-小鼠,6~8周龄,高脂饮食12周后,随机分为3组:对照组(Con组)、p53激动剂Parthenolide组(Par组)、p53抑制剂Pifithrin/-u组(PFT-u组),给药4周。处死动物后对主动脉油红O染色,颈总动脉茜素红染色、HE染色,主动脉血管功能实验。结果与对照组相比,p53抑制剂能显著减少动脉粥样硬化斑块面积、减少血管腔狭窄、抑制动脉内膜增生,改善胸主动脉血管功能。结论p53抑制剂可以抑制动脉粥样硬化斑块的形成。Objective To investigate the effect of p53 on the transdifferentiation from smooth muscle cells to osteoid cells in arterial calcification.Methods 21 healthy male apoE-/-mice were randomly divided into three groups after a high-fat diet fort 12 weeks,namely the control group,p53 agonist group and p53 inhibitor group.The drugs were injected intraperitoneally for 4 weeks.After the animals were sacrificed,the aorta was stained with oil red O,the common carotid artery was stained with alizarin red and HE,and the aorta was tested for vascular function.Results Compared with the control group,p53 inhibitor significantly reduced the area of atherosclerotic plaques,reduced vascular lumen stenosis,inhibited arterial intimal hyperplasia,and improved the vascular function of the thoracic aorta.Conclusion p53 inhibitors can inhibit the formation of atherosclerotic plaques.
