建鲤肠道中4种胞内酶和黏液细胞的分布及组织化学定位

摘　　要：为了研究酸性磷酸酶(ACP)、碱性磷酸酶(ALP)、过氧化物酶(POD)、非特异性酯酶(NSE)4种胞内酶及黏液细胞在建鲤Cyprinus carpio var.Jian(体质量为150 g±30 g)肠道组织中的分布特点,利用组织化学染色技术、光密度测定技术对其进行定量和定位分析。结果表明:ACP活性部位主要存在于肠上皮细胞核上区和固有层中,中肠ACP组织化学染色程度最强;ALP活性部位主要存在于肠道纹状缘和固有层,中肠ALP组织化学染色程度最强,前肠最低;POD活性部位主要存在于肠道固有层和黏膜肌层中,后肠POD组织化学染色程度最强;NSE活性在上皮细胞中尤为明显,后肠NSE组织化学染色程度最强;AB-PAS染色显示,肠道内有3种类型的黏液细胞(Ⅰ、Ⅱ、Ⅲ型),这3种黏液细胞在前肠中均分布在黏膜的上皮细胞中,在中肠和后肠中则分布在上皮和固有层中。研究表明,ACP、ALP、NSE、POD和黏液细胞广泛分布于建鲤的肠道组织中,且肠道不同部位的酶活性和黏液细胞分布具有不同特点。The distribution and location of acid phosphatase(ACP),alkaline phosphatase(ALP),peroxidase(POD),non-specific esterase(NSE)and mucous cells were qualitatively and quantitatively determined in the intestine of common carp Cyprinus carpio var.Jian with body mass of(150±30)g using enzyme histochemical,densitometry technology and statistical analyses.ACP activity was found in the supranuclear region of enterocytes and the lamina propria of the intestine,with the strongest staining in the middle intestine.ALP activity was detected in the striated border and lamina propria of the intestine,with the strongest staining(activity)of ALP in middle intestine and the minimal in the anterior intestine.POD activity was primarily found in the lamina propria and the mucosal muscularis of the intestinal,with the strongest staining in the posterior intestine.The activity of NSE was especially significant in epithelial cells and the staining intensity was strongest in the posterior intestine.Alcian blue(pH 2.5)periodic acid Schiff(AB-PAS)histochemical results revealed that in the intestines there were three types of mucous cells including typeⅠ,typeⅡ,and typeⅢ,which were distributed in the epithelial cells of the mucosa in the anterior intestine,and in the epithelium and lamina propria of the middle intestine and posterior intestine.In conclusion,the activities of ACP,ALP,NSE,and POD and mucous cells were generally distributed in the intestine of common carp,and the characteristics of the enzyme activity and mucous cell distribution were varied with different parts in the intestine.
