摘 要:为研究高原藏猪与约克夏猪PGM1基因多态性及mRNA水平的表达差异情况,实验以藏猪和约克夏猪作为实验动物,采用Sanger测序和PCR-RFLP法,对藏猪和约克夏猪5'侧翼区3000 bp序列进行多态性检测和基因分型;利用实时荧光定量PCR技术检测180日龄藏猪、约克夏猪PGM1基因在肝脏、背最长肌和背脂组织中mRNA水平相对表达量。结果发现:PGM1基因的5'侧翼区存在1个突变位点,且该位点符合哈迪-温伯格平衡,品种间表现差异极显著;PGM1基因在藏猪肝脏组织中的表达量极显著高于约克夏猪,在藏猪背最长肌组织中的表达量显著高于约克夏猪,在藏猪背脂组织中的表达量极显著低于约克夏猪。推测PGM1基因可能参与调控猪的肌肉生长发育和脂肪沉积,该结论为进一步研究猪的生长调控机制提供重要依据。In order to study the differences in PGM1 gene polymorphism and mRNA expression between plateau Tibetan pigs and Yorkshire pigs,the experiment used Tibetan pigs and Yorkshire pigs as experimental animals.The Sanger sequencing and PCR-RFLP method were used to compare Tibetan pigs and Yorkshire pigs.Pig 5'flanking region 3000bp sequence for polymorphism detection and genotyping;real-time fluorescent quantitative PCR technology was used to detect the relative expression of PGM1 genes in the liver,longissimus dorsi muscle and back fat tissue of 180-day-old Tibetan pigs and Yorkshire pigs.The results showed that there is a mutation site in the 5'flanking region of the PGM1 gene,and this site is in line with the Hardy-Weinberg equilibrium,showing extremely significant differences between breeds(P<0.01).The expression level of PGM1 gene in the liver tissues of Tibetan pigs was extremely significantly higher than that of Yorkshire pigs(P<0.01),and the expression level in the longissimus dorsi muscle tissue of Tibetan pigs was significantly higher than that of Yorkshire pigs(P<0.05).The expression level in pig back fat tissue was extremely significantly lower than that in Yorkshire pigs(P<0.01).In summary,it is speculated that the PGM1 gene may be involved in the regulation of muscle growth and fat deposition in pigs,providing an important basis for further research on the growth regulation mechanism of pigs.
