摘  要:目的利用生物信息学方法分析并预测细粒棘球绦虫肌醇多磷酸2-激酶(EgIP2K)的结构及抗原表位,为研究细粒棘球绦虫的药物靶点和疫苗开发提供理论依据。方法通过NCBI获取细粒棘球绦虫肌醇多磷酸2-激酶(EgIP2K)的氨基酸序列,采用Protparam预测EgIP2K的理化特性;采用SOPMA预测其二级结构;采用SignalP 4.1 server工具预测信号肽;采用NetPhos预测其磷酸化位点;采用DNAStar软件中的protean程序分析其抗原性指数、亲水性指数及表面可及性指数;采用ABCPred预测潜在的B细胞抗原表位;采用SYFPEITHI和IEDB工具预测蛋白的T细胞抗原表位。结果EgIP2K由492个氨基酸残基组成,相对分子质量为54.8×10^(3),分子式是C_(2397)H_(3791)N_(683)O_(730)S_(29),理论等电点为6.41,不稳定指数值为52.20,为不稳定蛋白;二级结构中α螺旋占36.10%,β折叠占16.46%,β转角占5.89%,无规则卷曲占46.54%;EgIP2K无信号肽序列,可能为非分泌蛋白;EgIP2K含有51个磷酸化位点,其中3个酪氨酸位点、13个苏氨酸位点、35个丝氨酸位点;预测EgIP2K具有B细胞和T细胞抗原表位,并且有3个抗原表位是B、T细胞联合抗原表位,分别位于90-94、411-422、424-438氨基酸区段。结论EgIP2K含有多个潜在的B细胞、T细胞抗原表位,可作为细粒棘球绦虫疫苗候选基因。Objective To use bioinformatics to examine the structure and potential epitopes of inositol polyphosphate 2-kinase of Echinococcus granulosus(EgIP2 K)in order to provide a theoretical basis to study the drug targets of and to develop vaccines against E.granulosus.Methods The amino acid sequence of EgIP2 K was downloaded from NCBI.The physicochemical properties of EgIP2 K were predicted using Protparam.Its secondary structure was predicted using SOPMA.SignalP 4.1 server was used to predict its signal peptides.Its phosphorylation sites were predicted using NetPhos,and analysis of its antigenic index,hydrophilicity,and surface probability was performed using the protean program in the software DNAStar.ABCPred was used to predict its potential B-cell epitopes,and SYFPEITHI and IEDB were used to predict its T-cell epitopes.Results EgIP2 K consisted of 492 amino acid residues with a molecular weight of 54.8×10^(3).Its molecular formula was C_(2397)H_(3791)N_(683)O_(730)S_(29)and its theoretical isoelectric point was 6.41.The instability index of EgIP2 K was 52.20,and thus it was considered to be an unstable protein.αhelices accounted for 36.10%of its secondary structure,βfolds accounted for 16.46%,βturns accounted for 5.89%,and random coils accounted for 46.54%.No signal peptide sequences were found in EgIP2 K,so it is likely to not be a secreted protein.The results of phosphorylation site prediction indicated that EgIP2 K had 51 phosphorylation sites,including 3 tyrosine sites,13 threonine sites,and 35 serine sites.EgIP2 K was predicted to have B-cell and T-cell epitopes.The protein has three potential B-cell and T-cell epitopes at amino acids 90-94,411-422,and 424-438.Conclusion With multiple potential B-cell and T-cell epitopes,EgIP2 K can serve as a potential gene for use in E.granulosus vaccines.
