摘 要:目的比较和评价脉冲场凝胶电泳技术(PFGE)和多位点可变数目串联重复序列分析技术(MLVA)在福建地区肠炎沙门菌分子分型中的应用价值。方法采用PFGE和MLVA技术对福建地区88株肠炎沙门菌分离株进行分子分型,结合流行病学资料分析比较分型效果。结果88株肠炎沙门菌经PFGE分型后,相似度系数为43.9%~100.0%,得到46个PFGE型别(PT 0001~PT 0046)、2个优势型别簇(cluster A~B),分辨系数(D值)为0.9459。PT 0002、PT 0011、PT 0013和PT 0029为优势PFGE;经MLVA分型后,遗传关联度介于26.8%~100%之间,得到40个MLVA型别(MT 0001~MT 0040)、2个优势基因簇(cluster 1~2),分辨系数(D值)为0.9235。MT 0005、MT 0004和MT 0014为优势MLVA型别。结论PFGE分辨力略高,与流行病学背景资料更吻合;MLVA在分析菌株之间亲缘关系和遗传相关性方面更具优势。Objective To compare and evaluate pulsed-field gel electrophoresis(PFGE) and multiple-locus variable-number tandem repeat analysis(MLVA) in the study of Salmonella Enteritidis in Fujian Province. Methods A total of 88 strains of Salmonella Enteritidis isolated from Fujian Province were genotyped using PFGE and MLVA, and the results were compared to epidemiological data. Results The similarity coefficient among the 88 isolates was from 43.9% to 100% according to PFGE, with a D index of 0.9459. Isolates were grouped into 46 PFGE types(TP0001-00046) with 4 predominant types(PT 0002, PT 0011, PT 0013, and PT 0029), and 2 dominant clusters(clusters A-B). MLVA resulted in 40 types, the degree of genetic association among isolates was from 26.8%-100%, with a D index of 0.9235. There were 3 predominant types(MT 0005, MT 0004, and MT 0014) and 2 dominant clusters(clusters 1-2). Conclusion PFGE has a slightly higher discriminatory ability compared to MLVA and its results are more consistent with the epidemiological data, but MLVA is better at analyzing genetic relationships and the genetic correlation between strains.
