摘 要:目的观察氧化槐果碱(OSC)对酒精诱导的小鼠急性肝损伤的保护作用。方法将50只昆明种小鼠随机分为5组,分别为正常对照组、模型组、OSC低、中、高剂量(10、20、40 mg·kg^(-1))组。OSC各剂量组采用灌胃方式提前预防给予OSC(10、20、40 mg·kg^(-1))2 d,2 d后用56°白酒灌胃(6 g·kg^(-1))以建立急性酒精性肝损伤模型。造模6 h后眼静脉丛取血,检测小鼠血清丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)的含量,肝组织超氧化物歧化酶(SOD)、过氧化氢酶(CAT)和谷胱甘肽过氧化物酶(GSH-Px)的活性以及丙二醛(MDA)的含量,观察小鼠体质量、肝脏指数和肝脏组织形态学结构的变化。结果各组小鼠初始体质量和终末体质量比较差异无统计学意义;不同剂量OSC(10、20、40 mg·kg^(-1))均可降低肝脏指数,减少血清肝损伤标志酶(ALT、AST)和肝组织脂质过氧化物(MDA)的含量,提高肝组织抗氧化酶(SOD、CAT和GSH-Px)的活性。组织病理学检查结果显示,OSC(10、20、40 mg·kg^(-1))可显著减轻小鼠肝脏组织病理学结构的异常改变。结论OSC对酒精致小鼠肝脏的结构和功能改变具有保护作用,其作用机制与OSC抑制酒精诱导的肝组织发生氧化应激、脂质过氧化和维持肝细胞膜稳定性有关。Objective To observe the protective effect of oxysophocarpine(OSC)on alcohol-induced acute liver injury in mice.Methods Fifty Kunming mice were randomly divided into the control group,the model group,and the OSC low,medium and high dose(10,20 and 40 mg·kg^(-1))groups.The OSC preventive administration groups were administered with OSC(10,20 and 40 mg·kg^(-1))by gavage for 2 days.After 2 days,the acute alcoholic liver injury model was established by gavage with 56°white wine(6 g·kg^(-1)),and the blood was taken from the ocular venous plexus after 6 hours of modeling.The serum levels of alanine aminotransferase(ALT)and aspartate aminotransferase(AST),the activity of superoxide dismutase(SOD),catalase(CAT)and glycine peroxidase(GSH-Px)and the content malondialdehyde(MDA)in liver tissues were detected,and the changes of body weight,liver index and liver tissue morphology of mice were observed.Results There was no significant difference between the initial body weight and the final body weight of mice in each group.Different doses of OSC(10,20 and 40 mg·kg^(-1))could reduce the liver index,the liver injury marker enzymes(ALT,AST)in serum and the lipid peroxide(MDA)content in liver tissue,and increase the activity of antioxidant enzymes(SOD,CAT and GSH-Px).The histopathological results showed that OSC(10,20 and 40 mg·kg^(-1))can significantly reduce the abnormal changes of the liver in mice.Conclusion OSC has a protective effect on alcohol-induced liver structure and function changes in mice,and its mechanism is related to the fact that OSC inhibits the alcohol-induced oxidative stress,lipid peroxidation in liver tissue,and maintains the liver cell membrane stability.
