摘 要:目的探讨桃花精颗粒(THJG)对小鼠糖尿病心肌病的影响及机制。方法采用高脂高糖饲料联合链脲佐菌素(STZ)建立小鼠糖尿病心肌病模型,建模成功后,随机分为5组,模型组及正常对照组给予等容量生理盐水,THJG各剂量组每天灌胃对应剂量药物,连续给药3个月。给药结束后,心脏超声检测小鼠心功能,随后取小鼠心脏称取质量并计算心脏指数,分离血浆,检测三酰甘油(TG)、高密度脂蛋白(HDL-C)、低密度脂蛋白(LDL-C)和超氧化物歧化酶(SOD)水平,血清用于检测白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、单核细胞趋化因子-1(MCP-1)和白细胞介素-10(IL-10)含量。结果高脂高糖饲料联合STZ造模小鼠的左心室射血分数(LVEF)和左心室短轴缩短率(LVFS)值分别为46.01%±8.51%、30.61%±5.16%,心脏指数显著增加至0.69%±0.08%,与正常对照组比较差异具有显著统计学意义(P<0.01),说明糖尿病心肌病建模成功。与模型组比较,THJG中、高剂量组的LVEF分别升高至58.48%±5.95%、60.12%±2.99%,LVFS分别升高至33.94%±4.85%、35.15%±3.03%,差异具有统计学意义(P<0.05或P<0.01),并呈明显的剂量依赖性;同时,明显降低心脏指数至0.64%±0.07%、0.63%±0.05%,降低血中TG、LDL-C、IL-6、TNF-α和MCP-1含量,升高HDL-C、SOD和IL-10含量(P<0.05或P<0.01)。结论THJG具有预防和治疗小鼠糖尿病心肌病的作用,其机制可能与提高小鼠抗氧化和抗炎作用相关。Objective To explore the effect and mechanism of Taohuajing Granules(THJG)on diabetic cardiomyopathy(DCM)in mice.Methods The DCM model was established by high-fat and high-sugar feed combined with streptozotocin(STZ).After the diabetes model was established,the mice were divided into 5 groups randomly.The control and model groups were given equal volume of saline,and the groups of THJG were administered with the corresponding dosage of the drug for 3 months consecutively.After the administration of the drug,cardiac ultrasonography was employed to detect the cardiac function of mice,and then the mouse heart was isolated and weighed to calculate the cardiac index.The plasma was separated to detect triglyceride(TG),high density lipoprotein(HDL-C),low density lipoprotein(LDL-C)and superoxide dismutase(SOD).The serum was used to detect the content of interleukin-6(IL-6),tumor necrosis factor-α(TNF-α),monocyte chemoattractant protein-1(MCP-1)and interleukin-10(IL-10).Results The left ventricular ejective fraction(LVEF)and left ventricular fractional shortening(LVFS)values of mice induced by high fat and high sugar diet combined with STZ were 46.01%±8.51%and 30.61%±5.16%,respectively,and the heart index was increased to 0.69%±0.08%,which was significantly different from that of normal group(P<0.01),indicating that the model of diabetic cardiomyopathy was successful.Compared with model group,the LVEF of THJG medium-dose and high-dose groups were increased to 58.48%±5.95%and 60.12%±2.99%,the LVFS were increased to 33.94%±4.85%,and 35.15%±3.03%,respectively,with statistically significant difference(P<0.05 or P<0.01)in a dose-dependent manner.At the same time,the cardiac index was significantly decreased to 0.64%±0.07%and 0.63%±0.05%,the contents of TG,LDL-C,IL-6,TNF-αand MCP-1 were decreased,and the levels of HDL-C,SOD and IL-10 were increased(P<0.05 or P<0.01).Conclusion THJG can treat diabetic myocardial injury in mice,and the mechanism may be related to the enhancement of antioxidant and anti-inflammatory ef
