摘 要:目的探究枸骨叶水提物(ICAE)对高脂血症大鼠血脂水平及抗氧化活性的影响。方法将72只健康的雄性SD大鼠分为空白对照组与模型组,空白对照组大鼠用基础饲料喂养,模型组大鼠用高脂饲料喂养构建高脂血症模型。将模型组大鼠分为模型对照组、阳性对照组(辛伐他汀,1.33 mg·kg^(-1))、ICAE低剂量(0.15 g·mL^(-1))组、ICAE中剂量(0.30 g·mL^(-1))组和ICAE高剂量(0.60 g·mL^(-1))组,空白对照组及模型对照组以生理盐水灌胃,其他组分别用相应药物灌胃,为期6周。每周称取大鼠体质量;检测大鼠血清指标和肝脏抗氧化指标;苏木精-伊红(HE)染色观察肝脏组织病理学形态。结果模型组大鼠与对照组相比体质量、总胆固醇(TC)、三酰甘油(TG)、低密度脂蛋白胆固醇(LDL-C)、动脉粥样硬化指数(AI 1和AI 2)、丙二醛(MDA)、肝脏指数显著增加,肝脏病变程度加深和脂肪沉积增加,而高密度脂蛋白胆固醇(HDL-C)、超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)显著降低(P<0.05);与模型对照组相比,阳性对照组及ICAE低、中和高剂量组大鼠的体质量、TC、TG、LDL-C、AI 1、AI 2、MDA和肝脏指数显著降低,肝脏病变程度和脂肪沉积减轻,而HDL-C、SOD和GSH-Px显著增加(P<0.05)。结论ICAE可有效降低高脂血症大鼠的血脂水平并提高大鼠肝脏抗氧化能力。Objective To Study the effects of Ilex cornuta aqueous extract(ICAE)on blood lipid level and antioxidant activity in hyperlipidemia rats.Methods A total of 72 healthy male SD rats were divided into control group and model group.The control group were fed with basal diet,while the model group was fed with high-fat diet.The model rats were divided into model control group,positive control(simvastatin group,1.33 mg·kg^(-1)),ICAE low-dose group(0.15 g·mL^(-1)),ICAE medium dose group(0.30 g·mL^(-1))and ICAE high-dose group(0.60 g·mL^(-1)).The control group and model group were given normal saline,and other treatment groups were given corresponding drugs by gavage,for 6 weeks.The rats were weighed every week;the serum and liver antioxidant indexes were detected;HE staining was used to observe the pathological morphology of liver.Results Compared with those in the control group,the body weight,total cholesterol(TC),triglyceride(TG),low density lipoprotein cholesterol(LDL-C),atherosclerosis indexes AI 1,AI 2,malondialdehyde(MDA),and liver index were significantly increased in the model group.The degree of liver lesion was deepened,and the fat deposition increased.However,the high density lipoprotein cholesterol(HDL-C),superoxide dismutase(SOD)and glutathione peroxidase(GSH-Px)were significantly decreased(P<0.05).Compared with the model group,the body weight,TC,TG,LDL-C,AI 1,AI 2,MDA and liver index were significantly decreased in the positive control group,low-dose group,middle dose group and high-dose group,the degree of liver lesions and fat deposition were reduced,and HDL-C,SOD and GSH-Px were significantly increased(P<0.05).Conclusion ICAE can effectively reduce the blood lipid level of hyperlipidemia rats and improve the antioxidant capacity of liver.
