摘  要:目的了解幽门螺杆菌(Helicobacter pylori,H.pylori)的免疫学检测技术和主要抗原表位及其应用,为临床诊断及生物技术药物开发提供参考。方法查阅相关文献并结合本课题组的研究,分析H.pylori的免疫学检测技术和主要抗原表位及其应用。结果基于抗原-抗体反应的免疫层析技术是H.pylori的主要体外检测技术。H.pylori特异性抗原是基于免疫反应H.pylori检测的关键,也是相关疫苗开发的关键。尿素酶(Ure)、细胞毒素相关蛋白A/L(CagA/L)、空泡毒素相关蛋白A(VacA)、中性粒细胞激活蛋白(NAP)是H.pylori致病性关键表面抗原和病理学基础。结论基于多种抗原(或抗体)的组合表位同时具备了高灵敏度和高特异性的优点,其在H.pylori诊断和疫苗及抗体等生物技术药物开发方面具有广阔的前景。Objective To review the immunological detection technology of Helicobacter pylori(H.pylori)and its main epitopes in order to provide reference for clinical diagnosis and vaccine development.Methods Based on the related literature and the research of our research group,the immunological detection technology of H.pylori,the main epitopes and their applications were analyzed.Results Rapid and low-invasive analysis of H.pylori infection is very important for the diagnosis and treatment of H.pylori infected gastropathy.H.pylori specific antigen is the key to the detection technology of H.pylori based on immune response,and also the key to the development of H.pylori vaccine.Urease(Ure),cytotoxin associated gene A/L(CagA/L),vacuolating toxin associated protein(VacA)and neutrophil activating protein(NAP)are the key surface antigens and pathlogical basis of H.pylori pathogenicity.Conclusion The combination epitopes based on multiple antigens(or antibodies)have the advantages of high sensitivity and specificity,showing a broad prospect in the diagnosis of H.pylori and the development of biotechnological drugs such as vaccines and antibodies.
