摘  要:为比较3处种源的西北小檗(Berberis vernae)的低温适应能力,文中以青海高原分布的3处种源的西北小檗150 d生幼苗为研究对象,测定不同种源枝条的低温半致死温度;同时人工控温进行低温胁迫,测定不同种源西北小檗幼苗叶片中MDA、可溶性蛋白和可溶性糖的含量以及SOD、POD和CAT活性的变化规律。结果显示:3处种源的西北小檗枝条低温半致死温度由低到高的顺序为玉树市下拉秀乡-21.55℃、西宁市大通县-16.83℃、海东市乐都区-15.78℃;低温条件下,玉树市下拉秀乡种源的西北小檗体内可溶性糖含量、可溶性蛋白含量、SOD活性、POD活性和CAT活性的增加量均要高于西宁市大通县种源和海东市乐都区种源,说明玉树市下拉秀乡种源的低温适应能力较其他两处种源强。本研究结果可为西北小檗耐寒种质的筛选、耐寒基因的挖掘以及高海拔城镇绿化树种的种源地选择提供理论依据。In order to compare the low temperature adaptability of Berberis vernae from three provenances,the 150-day seedlings of Berberis vernae from three provenances in Qinghai plateau are studied to determine the low-temperature semi-lethal temperature of branches from different provenances;at the same time,the content of MDA,soluble protein and soluble sugar are measured under the stress of low temperature as well as the activities of SOD,POD and CAT in the leaves of Berberis vernae seedlings from different provenances.The results show that the order of low-temperature semi-lethal temperature of Berberis vernae branches from three provenances is Yushu Xialaxiu-21.55℃,Xining Datong-16.83℃and Haidong Ledu-15.78℃.The increase of soluble sugar,soluble protein,SOD activity,POD activity and CAT activity in Berberis vernae from Yushu Xialaxiu are higher than those from Xining Datong and Haidong Ledu under low temperature,indicating that Yushu Xialaxiu has stronger adaptability to low temperature than the other two provenances.The results can provide a theoretical basis for the screening of cold resistant germplasm,the mining of cold resistant genes and selection of provenance of high-altitude urban greening tree species.
