补精益视片对SD大鼠慢性EIOP模型LGN中PI3K/Akt通路内GSK3β、CREB表达的影响

摘　　要：目的观察补精益视片对SD大鼠慢性高眼压(elevated intraocular pressure, EIOP)模型外侧膝状体(lateral geniculate nucleus, LGN)中PI3K/Akt通路内GSK3β及CREB表达的影响,并探讨其作用机制。方法采用烙闭上巩膜静脉法,建立SD大鼠慢性EIOP模型,30只SD大鼠随机分为对照组、模型组及治疗组。连续灌胃8周,并于8周末处死大鼠,观察SD大鼠慢性EIOP模型眼压、LGN尼氏小体含量、PI3K/Akt通路GSK3β及CREB表达。结果 GSK3β:模型组面积总和、平均黑度和积分光密度总和最高,与对照组、治疗组相比,差异均有统计学意义(P<0.05)。与对照组相比,治疗组面积总和、积分光密度总和均稍高,但差异无统计学意义(P>0.05);治疗组平均黑度较高,差异均有统计学意义(P<0.05)。CREB:对照组面积总和、平均黑度和积分光密度总和最高,与模型组、治疗组相比,差异均有统计学意义(P<0.05)。与模型组相比,治疗组面积总和、平均黑度和积分光密度总和均较高,差异有统计学意义(P<0.05)。结论补精益视片可有效保护SD大鼠慢性EIOP模型的视功能,其作用表现在下调LGN中GSK3β表达,上调LGN中CREB的表达。Objective To observe the effect of Bujingyishi Tablet on the change of Nissl body volume in LGN and the expression of GSK3β and CREB in PI3 K/Akt pathway in SD rats. Methods The chronic EIOP model of SD rats was established by the method of sealing the upper scleral vein. 30 SD rats were randomly divided into control group, model group and treatment group. The rats were sacrificed for 8 weeks, and the rats were sacrificed at the end of 8 weeks. The intraocular pressure, LGN Nissl body content, PI3 K/Akt pathway GSK3β and CREB expression in SD rats were observed. Results①GSK3β:The sum of the model group, the mean blackness mean and the integrated optical density were the highest.Compared with the control group and the treatment group, the difference was statistically significant(P<0.05).Compared with the control group, the total area and integrated optical density of the treatment group were slightly higher, but the difference was not statistically significant(P>0.05).The mean blackness of the treatment group was higher, the difference was statistically significant(P<0.05).).②CREB:The sum of the area of the control group, the mean blackness and the total integrated optical density were the highest.Compared with the model group and the treatment group, the difference was statistically significant(P<0.05).Compared with the model group, the total area, mean blackness and integrated optical density of the treatment group were higher, and the difference was statistically significant(P<0.05).Conclusion Bujingyishi tablets can effectively protect the visual function of the chronic EIOP model of SD rats. Its effect is manifested in down-regulating the expression of GSK3β in LGN and up-regulating the expression of CREB in LGN.
