摘  要:目的探索血塞通软胶囊(三七总皂苷)对脑出血大鼠脑组织的保护作用。方法用胶原酶注射尾状核法构建脑出血大鼠模型,并随机分为对照组和实验组,每组50只;另取50只正常大鼠作为假手术组。假手术组与对照组均灌胃给予等量0.9%NaCL;实验组按50 mg·kg^(-1)·d^(-1)的剂量灌胃给予100 mg·mL^(-1)三七总皂苷(血塞通软胶囊)溶液。3组大鼠每天给药1次,连续给药28 d。用干-湿重法检测脑组织含水量,用分光光度仪法检测脑组织一氧化氮(NO)、丙二醛(MDA)含量和超氧化物歧化酶(SOD)活性。结果治疗28 d后,实验组、对照组和假手术组脑组织的含水量分别为(78.89±1.57)%,(82.39±1.70)%和(77.83±1.57)%,NO分别为(0.50±0.11),(0.63±0.13)和(0.19±0.10)μmol·g-1,MDA分别为(13.80±1.19),(15.86±1.42)和(9.92±1.37)nmol·mg^(-1),SOD活性分别为(197.58±11.85),(157.39±16.51)和(199.99±10.81)U·mg^(-1);实验组的上述指标与对照组比较,差异均有统计学意义(均P<0.05)。结论血塞通软胶囊(三七总皂苷)可通过抑制自由基损伤和氧化应激,缓解脑水肿,从而达到保护脑出血大鼠脑组织的作用。Objective To explore the protective effect of Xuesaitong soft capsules (Panax notoginseng total saponin) on brain tissue of rats with cerebral hemorrhage.Methods Rat model of cerebral hemorrhage was constructed by collagen enzyme injection,and randomly divided into control and experimental groups with 50 cases per group.Another 50normal rats were taken as sham-operation group.The sham-operation and control groups were gavaged the equal amount of 0.9%NaCl.The experimental group was given 100 mg·m L;Panax notoginseng total saponin (Xuesaitong soft capsule) solution by gavage at a dose of 50mg·kg;·d;.Three groups were administered once a day for 28 d.The water content of brain tissue was measured by the dry-wet weight method.The nitric oxide (NO) and malondialdehyde (MDA) contents,and superoxide dismutase (SOD) activity of brain tissue were measured by the spectrophotometric method.Results After 28 d of treatment,the water contents of hindbrain tissue in experimental,control and shamoperation groups were (78.89±1.57)%,(82.39±1.70)%and(77.83±1.57)%,brain tissue NO levels were (0.50±0.11),(0.63±0.13) and (0.19±0.10)μmol·g;,MDA levels were (13.80±1.19),(15.86±1.42) and(9.92±1.37) nmol·mg;,SOD activities were (197.58±11.85),(157.39±16.51) and (199.99±10.81) U·mg;.The above indicators in the experimental group were statistically significant when compared with the control group (all P<0.05).Conclusion Xuesaitong soft capsules (Panax notoginseng total saponin) can protect brain tissue in rats with cerebral hemorrhage by inhibiting free radical damage and oxidative stress and alleviating brain edema.
