摘  要:目的:观察瘢痕疙瘩组织中微小RNA-22-5p(miR-22-5p)和真核细胞翻译起始因子4E(eIF4E)的表达,分析其相关性及临床意义。方法:选择51例瘢痕疙瘩组织作为观察组,51例增生性瘢痕组织作为对照组,51例正常皮肤组织作为正常对照组。应用实时荧光定量PCR(qRT-PCR)法检测三组中miR-22-5p的表达,应用免疫组化法和Western Blot法检测瘢痕疙瘩组织中eIF4E的表达。结果:观察组中miR-22-5p的表达明显低于对照组和正常对照组,差异有统计学意义(P<0.05)。miR-22-5p的表达在不同病变最大径、增殖指数和有无家族史中的表达差异有统计学意义(P<0.05);而在不同性别、年龄中的表达差异无统计学意义(P>0.05)。瘢痕疙瘩中miR-22-5p与eIF4E呈负相关。结论:MiR-22-5p在瘢痕疙瘩中的表达下降,与临床及病理特征有关,miR-22-5p与eIF4E可能具有协同负向作用。Objective To observe the expression of microRNA-22-5p(miR-22-5p)and eukaryotic translation initiation factor 4E(eIF4E)in keloid tissue,and analyze their correlation and clinical significance.Methods 51 cases of keloid tissue were selected as the observation group,51 cases of hypertrophic scar tissue were selected as the control group,51 cases of normal skin tissue were selected as the normal control group.The expression of miR-22-5p was detected by real-time quantitative PCR(qRT-PCR)in three groups The expression of eIF4E was detected by immunohistochemistry and Western blot in keloid tissue.Results The expression of miR-22-5p in the observation group was significantly lower than that in the control group and the normal control group,the differences were statistically significant(P<0.05).There were significant differences in the expression of mir-22-5p in the maximum diameter of different lesions,proliferation index and family history(P<0.05).However,there was no significant difference in expression between different genders and ages(P>0.05).Negative correlation was found between miR-22-5p and eIF4E in keloid.Conclusion The expression of miR-22-5p is decreased in keloid,which is related to the clinical and pathological characteristics.MiR-22-5p and eIF4E may have synergistic negative effect.
