知百安神口服液抗抑郁作用及机制研究

摘　　要：目的:探讨知百安神口服液的抗抑郁作用及作用机制。方法:通过小鼠绝望模型和利血平拮抗抑郁模型,观察知百安神口服液对小鼠抑郁的改善作用;按单胺假说理论,观察知百安神口服液抑制单胺类递质重摄取和抑制单胺氧化酶的作用。结果:知百安神口服液能明显缩短小鼠悬尾和强迫游泳不动时间,明显降低小鼠眼睑下垂度,提示知百安神口服液具有抗抑郁作用。机制研究结果表明,知百安神口服液对多巴胺重摄取的抑制作用不明显;对5-羟色胺、去甲肾上腺素的重摄取有明显的抑制作用;对盐酸色胺引起的大鼠前肢痉挛行为有明显的增强作用。结论:知百安神口服液具有抗抑郁作用,其机制可能与增强脑内5-羟色胺、去甲肾上腺素神经递质作用以及抑制单胺氧化酶有关。OBJECTIVE:To probe into the antidepressant function and mechanism of Zhibai Anshen oral liquid.METHODS:The effect of Zhibai Anshen oral liquid on the improvement of depression in mice were observed by the mouse despair model and the reserpine antagonistic depression model;the effect of Zhibai Anshen oral liquid in inhibiting the monoamine oxidase and reuptake of monoamine transmitters were observed according to the monoamine hypothesis theory.RESULTS:Zhibai Anshen oral liquid can significantly shorten the duration of tail suspension and forced swimming immobility in mice,and significantly reduced the eyelid ptosis in mice,which suggested that Zhibai Anshen oral liquid had antidepressant effect.The results of mechanism study showed that Zhibai Anshen oral liquid had no significant inhibitory effect on the reuptake of dopamine;while it had significant inhibitory effect on the reuptake 5-hydroxytryptamine and norepinephrine;and had significant enhancement on the forelimb spastic behavior of rats induced by tryptamine hydrochloride.CONCLUSIONS:Zhibai Anshen oral liquid has antidepressant effect,and the mechanism may be related to the enhancement of 5-hydroxytryptamine and norepinephrine neurotransmitter effect in brain and the inhibition of monoamine oxidase.
