摘  要:[目的]建立番茄抗斑萎病毒病基因Sw-5的分子标记鉴定技术,提高抗病育种材料的选择效率。[方法]利用PCR技术和6份(3份抗病和3份感病)的番茄自交系,对获得的与Sw-5紧密连锁的分子标记进行验证,建立稳定可靠的PCR反应体系。[结果]通过验证,纯合抗病材料(Sw-5/Sw-5)扩增出574 bp条带,感病材料(Sw-5/Sw-5)扩增出464 bp条带;利用该标记对8份抗病Fi杂交种进行鉴定,5份材料为杂合抗病(sw-5/sw-5),扩增出574 bp和464 bp两条带,1份材料为纯合抗病,2份材料未检出Sw-5基因;对抗病杂交种的28份F_(2)代分离单株进行鉴定,获得纯合抗病单株6份,杂合抗病单株14份。[结论]该标记是与Sw-5基因紧密连锁的共显性标记,稳定可靠,可用于番茄抗病育种材料的鉴定和筛选。[Objective] The aim was to develop a molecular marker for assisted-selection of the Sw-5(Tomato spotted wilt virus resistance) gene,which would increase the selection efficiency of resistant materials.[Method] Six tomato inbred lines including 3 resistant and 3 susceptible materials were selected to test the molecular marker which was tightly linked to Sw-5 by using PCR technology,and a stable and reliable PCR reaction system was established. [Result] The PCR results showed that resistant and susceptible homozygous materials amplified 574 bp and 464 bp bands respectively;Eight resistant F_(1) hybrids were detected with this marker. The results showed that 5 materials contained heterozygous Sw-5 gene,and amplified 574 bp and 464 bp bands,1 resistant homozygous material,2 materials didn’t contain Sw-5 gene;The SCAR marker was also used to detect 28 F_(2) individuals. The results showed that 6 individuals contained homozygous Sw-5 gene,14 individuals contained heterozygous Sw-5 gene. [Conclusion] The SCAR was a co-dominant marker which tightly linked to Sw-5 gene,and the results are stable and reliable,this marker could be used for identification and screening of tomato resistant breeding materials.
