摘 要:目的:建立一测多评法进行九制大黄丸中5种蒽醌类成分的含量测定。方法:以大黄素为内参物,建立了芦荟大黄素等4个其他游离型蒽醌与大黄素的相对校正因子和待测成分的色谱峰定位方法,并进行方法学考察。在此基础上,分别采用外标法测定和一测多评法计算10批九制大黄丸中5个成分的含量,比较实测值与计算值的差异,以验证一测多评法的准确性。结果:建立的相对校正因子重现性良好,色谱峰相对保留值可用于待测成分色谱峰的定位,芦荟大黄素、大黄酸、大黄素、大黄酚、大黄素甲醚5个成分采用校正因子计算的含量值与实测值之间差异均无统计学意义。结论:一测多评法进行九制大黄丸中5种蒽醌类成分的含量测定,方法可行。Objective:To establish a evaluation method for content determination of five anthraquinones in Jiuzhi Dahuang Wan by quantitative analysis of multi-components by a single marker.Methods:Emodin was used as internal reference,the relative correction factor of 4 other free anthraquinones and emodin were established,and the chromatographic peak localization method of the components to be measured.We conducted a methodological survey.On this basis,the contents of 5 components in 10 batches of rhubarb pills were calculated by external standard method and quantitative anaiysis of multi-components by a single marker,to compare the difference between measured value and calculated value for verify the accuracy of the multi-evaluation method.Results:The relative correction factor established has good reproducibility.The relative retention value of chromatographic peak can be used to locate the chromatographic peak of the component to be measured.The contents of aloe emodin,rhein acid,emodin,chrysophanol and emodin methyl ether were not significantly different fromthose calculated by correction factor.Conclusion:The way to content determination of fiveanthraquinones in Jiuzhi Dahuang Wan byquantitative analysis multi-components by single markers is useful.
