摘 要:目的研究陈皮95%乙醇提取物对脂肪乳剂(“过食肥腻”)致高脂血症大鼠模型的改善作用。方法SD大鼠随机分为正常组、模型组、阳性对照组及陈皮95%乙醇提取物高、中、低剂量组,每组8只,除正常组外,其余各组灌胃相应体积脂肪乳剂。造模同时各给药组灌胃相应体积药物,正常组和模型组灌胃等量生理盐水,每天1次,连续给药4周,自由饮食饮水。实验结束前,MoorFLPI散斑全帧实时扫描成像系统检测大鼠微循环血流量。实验结束后取血、取材,计算脏器系数,观察脏器外观,检测血清中总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-c)、天冬氨酸氨基转移酶(AST)、谷丙转氨酶(ALT)、肌酐(Cr)、尿酸(UA)等指标,酶联免疫吸附(ELISA)法检测血清中载脂蛋白A1(ApoA1)、载脂蛋白B(ApoB)、羟甲基戊二酰辅酶A还原酶(HMGCR)、胆固醇7α羟化酶(CYP7α1)、酰基辅酶A-胆固醇酰基转移酶(ACAT)、卵磷脂胆固醇脂酰转移酶(LCAT)水平。结果与模型组比较,陈皮醇提物组TC、LDL-c、ApoB水平下降(P<0.01),ApoA1水平升高(P<0.01),ALT、AST活性和UA水平降低(P<0.05),HMGCR活性降低(P<0.01),同时改善微循环血流量和主要脏器肿大现象。结论陈皮具有明显的降血脂作用,其作用机制可能是通过调控HMGCR活性从而减少胆固醇的内源性合成,同时可调控载脂蛋白(ApoA1和ApoB)的水平而避免血清TC的过量蓄积。AIM To study the effect of 95%ethanol extract of Citri Reticulatae Pericarpium on a rat model of hyperlipidemia induced by fat emulsion(high-cholesterol diet).METHODS SD rats were randomly divided into normal group,model group,positive control group,groups for interventions of high dose,medium dose and low dose of 95%ethanol extract of Citri Reticulatae Pericarpium,with eight rats in each group.Except for the normal group,the other groups were simultaneously given the corresponding volume of fat emulsion and the corresponding volume of drugs or normal saline once daily for four consecutive weeks,given diversified diet and water.Prior to the end of the experiment,the MoorFLPI speckle full-frame real-time scanning imaging system was used to detect the blood flow of the rat microcirculation.After the experiment,the rats procured with blood and other materials were subjected to the calculation of the organ coefficient,the observation of the organ appearances,the detections of the total cholesterol(TC),low-density lipoprotein cholesterol(LDL-c),aspartate aminotransferase(AST),gluten alanine aminotransferase(ALT),creatinine(Cr),uric acid(UA)and other indicators,and the determination of the levels of serum apolipoprotein A1(ApoA1),apolipoprotein B(ApoB),hydroxymethylglutaryl-coenzyme A reductase(HMGCR),cholesterol 7αhydroxylase(CYP7α1),acyl-CoA-cholesterol acyltransferase(ACAT)and lecithin cholesterol acyltransferase(LCAT)by ELISA method.RESULTS Compared with the model group,the groups intervened with the alcohol extract of Citri Reticulatae Pericarpium shared decreased levels of TC,LDL-c,and ApoB(P<0.01),increased levels of ApoA1(P<0.01);decreased activities of ALT and AST,the levels of UA(P<0.05)and HMGCR(P<0.01);in addition to the improvements in the microcirculation blood flow and the swelling of the main organs.CONCLUSION The significant effects of Citri Reticulatae Pericarpium in lowering blood lipids may contribute to its efficacy in regulating the activity of HMGCR to reduce the endogenous synthesis of cholest
