摘  要:动脉粥样硬化是一种炎症引起的慢性疾病,以巨噬细胞脂质代谢功能障碍导致的泡沫细胞形成为主要标志。载脂蛋白A(ApoA)-Ⅰ是高密度脂蛋白的重要结构蛋白组分,在机体脂质代谢过程中具有介导泡沫细胞胆固醇外流、维持机体胆固醇稳态、抗动脉粥样硬化的作用。此外,ApoA-Ⅰ作为一种多功能蛋白,在胆固醇逆向转运过程中起重要作用。ApoA-Ⅰ及其模拟肽可增强泡沫细胞胆固醇流出,具有抗动脉粥样硬化、抗炎、抗氧化等作用,在动脉粥样硬化性心血管疾病的防治方面具有很大的应用潜力。因此,对ApoA-Ⅰ及其模拟肽介导胆固醇外流的功能和特性的研究可为抗动脉粥样硬化药物的开发提供新思路。Atherosclerosis is a chronic disease caused by inflammation,and the marker is the formation of foam cells due to the dysfunction of macrophage lipid metabolism.Apolipoprotein A(ApoA)-Ⅰ is a major protein component of high-density lipoprotein,which mediates cholesterol efflux from macrophage-derived foam cells and maintains cholesterol homeostasis,preventing atherosclerosis.In addition,ApoA-Ⅰ as a multifunctional protein which plays an important role in reverse cholesterol transport(RCT).ApoA-Ⅰ and its mimetic peptides may promote cholesterol efflux from foam cells and have the anti-atherosclerosis,anti-inflammatory and anti-oxidant effects,showing great potential in prevention of atherosclerotic cardiovascular disease.Therefore,understanding the properties and functions of ApoA-Ⅰand its mimetic peptides mediated cholesterol efflux may provide some new strategies for the drugs development of anti-atherosclerosis.
