摘 要:目的:通过观察单次大剂量电子线照射SD大鼠正常胰腺的病理改变、核内因子-κB(NF-κB)和白细胞介素-10(IL-10)随时间变化的情况,探讨其在放射性损伤中的作用。方法:利用专利技术“小动物放射线精确照射试验台”以8MeV电子线20Gy单次准确照射SD大鼠胰腺,通过HE染色观察胰腺组织的病理学变化,采用免疫组织化学染色观察NF-κB和IL-10蛋白的表达。结果:照射后2周、4周胰腺组织细胞以水肿、坏死为主,伴炎症细胞浸润;6周、8周萎缩的胰腺细胞减少,胰岛细胞周围见大量纤维组织。胰腺中NF-κB和IL-10在照射后2周、4周的表达呈进行性增加,于6周、8周表达逐渐下降,各实验组均高于对照组(均P<0.05);照射后NF-κB与IL-10在胰腺中的表达呈正相关(r=0.914,P<0.05)。结论:单次大剂量电子线照射SD大鼠后,胰腺组织早期有一过性损伤,随后出现代偿性修复,可能与抗炎因子IL-10表达升高,抑制NF-κB的过度表达有关。Objective:To observe the pathological changes of normal pancreas,nuclear factor-κB(NF-κB)and interleukin-10(IL-10)over time in SD rats by single high-dose electron ray irradiation,explore its role in radiation damage.Methods:Using the patented technology"small animal radiation precise irradiation test bench"to irradiate SD rat pancreas with 8 MeV electron ray 20 Gy,the pathological changes of pancreatic tissue were observed by HE staining,the expression of NF-κB and IL-10 protein was observed by immunohistochemical staining.Results:After 2 weeks and 4 weeks of irradiation,pancreatic tissue cells were mainly edema and necrosis,accompanied by inflammatory cell infiltration;6 weeks and 8 weeks atrophy of pancreatic cells decreased,and a lot of fibrous tissue was seen around the islet cells.The expression of NF-κB and IL-10 in the pancreas increased progressively at 2 and 4 weeks after irradiation,and gradually decreased at 6 and 8 weeks,and the experimental groups were higher than those in the control group(all P<0.05):There was a positive correlation between the expression of NF-κB and IL-10 in the pancreas(r=0.914,P<0.05).Conclusion:After a single high-dose electron ray irradiation in SD rats,the pancreatic tissue was temporarily damaged in the early stage,and then compensatory repair occurred,which may be related to the increased expression of anti-inflammatory factor IL-10 and the inhibition of NF-κB overexpression.
