摘 要:目的探讨重组脂联素对自然流产模型小鼠子宫蜕膜组织中FOXO3蛋白表达的影响。方法将24只自然流产模型小鼠按照随机数字表法分为3组—模型组、脂联素1组、脂联素2组,每组8只。3组分别给予小鼠腹腔注射重组脂联素,剂量分别为0、0.1、1 mg/kg,1次/d,连续应用14 d。治疗第7天与治疗第14天,观察胚胎情况,计数吸收胚胎数和存活胚胎数;检测脂联素、白细胞介素-17(IL-17)、转化生长因子-β(TGF-β)等表达水平;检测脾脏CD4+CD25+T细胞比率;检测子宫蜕膜组织中FOXO3蛋白表达变化情况。结果治疗第7天与治疗第14天,脂联素1组、脂联素2组的胚胎吸收率均低于模型组,差异均有统计学意义(P<0.05),脂联素2组低于脂联素1组,差异均有统计学意义(P<0.05)。治疗第7天与治疗第14天,脂联素1组、脂联素2组的血清脂联素、TGF-β水平高于模型组,血清IL-17水平低于模型组,脂联素2组与脂联素1组对比,差异均有统计学意义(P<0.05)。治疗第7天与治疗第14天,脂联素1组、脂联素2组的脾脏CD4+CD25+T细胞比率高于模型组(P<0.05),脂联素2组高于脂联素1组,差异均有统计学意义(P<0.05)。治疗第7天与治疗第14天,脂联素1组、脂联素2组的子宫脱膜组织FOXO3蛋白相对表达水平高于模型组(P<0.05),脂联素2组高于脂联素1组,差异均有统计学意义(P<0.05)。结论重组脂联素在自然流产模型小鼠的应用能促进子宫蜕膜组织中FOXO3蛋白表达,提高脾脏CD4^(+)CD25^(+)T细胞比率,调节小鼠的免疫平衡,从而能降低胚胎吸收率。Objective To investigate the effect of recombinant adiponectin on FOXO3 protein expression in decidua tissue of spontaneous abortion model mice.Methods The spontaneous abortion model 24 mice were randomly divided into three groups-model group,adiponectin 1 group,and adiponectin 2 group according to the random number table method,8 mice in each group.The three groups were given intraperitoneal injection of recombinant adiponectin in the mice,and the doses were as follows:0,0.1,1 mg/kg,1 time/d,applied 14 d continuously.At the 7th and 14th day of treatment,the condition of embryos was observed,the number of absorbed embryos and the number of surviving embryos were calculated;the expression levels of adiponectin,interleukin-17(IL-17),transforming growth factor-β(TGF-β),etc.The ratio of CD4+CD25+T cells in the spleen was tested;the expression of FOXO3 protein in the decidua tissue of the uterus was tested.All the mice were to detect the changes in FOXO3 protein expression in decidua tissue.Results At the 7th and 14th day of treatment,the embryo absorption rate in adiponectin 1 group and adiponectin 2 group were lower than those in the model group,the differences were statistically significant(P<0.05),and adiponectin 2 group were lower than those in adiponectin 1 group,the differences were statistically significant(P<0.05).At the 7th and 14th day of treatment,the serum adiponectin and TGF-β1 levels in adiponectin 1 group and adiponectin 2 group were higher than those in the model group,and the serum IL-17 level were lower than those in the model group(P<0.05),compared the difference between adiponectin 2 group and adiponectin 1 group were also statistically significant,the differences were statistically significant(P<0.05).At the 7th and 14th day of treatment,the proportion of CD4+CD25+T cells of the spleen in adiponectin 1 group and adiponectin 2 group were higher than those in the model group(P<0.05),and adiponectin 2 group were higher than adiponectin 1 group,the differences were statistically significant(P<0.05
