摘 要:目的优选大孔树脂与无机陶瓷膜联用精制消肿止痛液的工艺。方法以盐酸小檗碱吸附容量、洗脱率为评价指标,通过静态吸附-洗脱试验从AB-8、HPD600、D101三种型号树脂优选出大孔树脂型号,通过正交试验考察上样浓度、大孔树脂用量、乙醇体积分数及体积对盐酸小檗碱得率的影响;以盐酸小檗碱转移率为评价指标,衡量不同孔径无机陶瓷膜及运行参数对水提液的精制效果。结果AB-8型大孔吸附树脂对盐酸小檗碱的吸附和洗脱性能均较好,最佳上样浓度为1.0 g·mL^(-1),大孔树脂用量(g)与上样量(mL)比为8∶1,60%乙醇洗脱剂浓度,洗脱剂(mL)与上样量(mL)比为8∶1。选用膜孔径为800 nm的无机陶瓷膜,压差0.2 MPa,过膜温度40℃,药液浓度为0.1 g·mL^(-1)条件下,膜通量及盐酸小檗碱的转移率均较高。结论该试验表明大孔树脂与无机陶瓷膜联合用于消肿止痛液中盐酸小檗碱精制方法可行。Objective To optimize the purification technology of Xiaozhong Zhitong Lotion by Macroporous Adsorptive Resin combined with Inorganic Ceramic Membrane.Methods Taking the adsorption capacity and elution rate of berberine hydrochloride as evaluation indexes,the macroporous resin models were selected from AB-8,HPD600 and D101 through static adsorption elution test.The effects of sample concentration,macroporous resin dosage,ethanol volume fraction and volume on the yield of berberine hydrochloride were investigated by orthogonal test.Taking the transfer rate of berberine hydrochloride as the evaluation index,the refining effect of inorganic ceramic membrane with different pore sizes and operating parameters on water extract was measured.Results The result showed that AB-8 macroporous adsorptive resin had a better adsorption and desorption properties than the others.The best concentration of sample solution was 1.0 g·m L^(-1),the ratio of wet resin amount(g)and sample volume(mL)was 8∶1,60%alcohol as the eluent,and the ratio of eluant and loading amount was 8∶1.The membrane flux and metastasis rates of berberine hydrochloride were higher under the condition of pore size of 800 nm,0.2 MPa of operation pressure,40℃of filtration membrane temperature and concentration of 0.1 g·m L^(-1).Conclusion It was feasible to use macroporous adsorptive resin and inorganic ceramic membrane to improve the internal quality of berberine hydrochloride in Xiaozhong Zhitong Lotion.
