摘  要:天麻素是我国名贵中药材天麻中的一种重要的药效组分,具有降血压、抑制肿瘤等重要的功能,然而,天麻素的生物合成途径尚有待于揭示。本研究以天麻基因组、转录组数据进行分析,转录组序列组装后获得48 823个转录本,平均长度为2 163 nt, N50长度为2 689 nt,GC含量为49.42%, 2 000 bp以上所占比例最大,共21 608个(44.26%),大于1 000 bp共有39 007个(79.89%)。对unigene进行KEGG注释分析,共17 629个(注释率36.11%) unigene获得功能注释,映射到274条分支代谢途径中。其中,与天麻素生物合成密切有关的苯丙酸合成和苯丙氨酸代谢的unigene分别为248和119个。通过共表达分析,表明天麻素合成底物4-羟基苯甲醇可能主要来源于苯丙氨酸代谢途径。该研究将为后续基因功能研究及天麻药效品质研究提供理论依据和实验参考。Gastrodin is an important component of Gastrodia elata in China, which has the functions of lowering blood pressure and inhibiting tumor. However, the biosynthetic pathway of gastrodin remains to be revealed. In this study, a total of 48 823 assembled unigenes were obtained, the average unigenes length is 2 163 nt, 21,608 unigenes(44.26%) are more than 2 000 bp, and 39 007 unigenes(79.89%) are more than 1 000 bp. The length of N50 is 2 689 nt, and the GC content is 49.42%. Analyzing the KEGG metabolic pathway of the unigenes, 17 629(36.11%) unigenes obtained KEGG functional annotation in total of 274 KEGG metabolic pathways, of which 248 unigenes participate in phenylpropanoid biosynthesis, and 119 unigenes participate in phenylalanine metabolism which involving gastrodin biosynthesis. Through co-expression analysis, it is shown that the 4-hydroxybenzyl alcohol may mainly come from phenylalanine metabolism. This study will provide a theoretical basis and experimental reference for the study of molecular mechanism of quality of G. elata.
