摘  要:目的:探讨疏肝理脾汤对肝郁脾虚证非酒精性脂肪性肝炎(NASH)大鼠粪便菌群多样性和丰富度的影响。方法:采用蛋氨酸胆碱缺乏(MCD)饮食联合慢性束缚应激刺激法复制NASH大鼠,造模成功后,将大鼠随机分为模型组、疏肝理脾高剂量组、疏肝理脾低剂量组,同时选取同批次非造模大鼠为正常对照组,不予药物干预。进入实验观察阶段后,疏肝理脾高、低剂量组大鼠分别给予相应药物灌胃治疗,模型组和正常对照组大鼠均给予等量蒸馏水灌胃。实验结束后,行16srRNA肠道菌群多样性检测。结果:肠道菌群OUTs值检测结果提示,正常对照组和NASH大鼠菌群种类门水平优势菌群种类无变化,但属水平部分模型组与疏肝理脾高剂量组比较,菌群种类比例有变化,有18个菌属为疏肝理脾高剂量组特有,有7个菌属为模型组特有。Beta多样性检测模型组、疏肝理脾高剂量组组间差异,提示两组间Bray crutis和unweight unifrac均较远且无交集,提示两者之间在物种丰富度上差异较大。LEfSe(LDA Effect SiSe)、热图分析模型组、疏肝理脾高剂量组两组间的特征性差异菌,提示模型组与疏肝理脾高剂量组大鼠肠道菌群的丰度存在明显差异。结论:疏肝理脾汤能改善肝郁脾虚证NASH大鼠肠道菌群物种丰富度及物种之间占比,可通过降低普氏菌科、毛螺菌科,提高梭菌科、拟杆菌科,从而调节肠道菌群构成比,影响肠道菌群属水平,恢复肠道微生态环境平衡。Objective:To observe the effect of Shugan Lipi Decoction on the diversity and richness of fecal flora in rats with nonalcoholic steatohepatitis(NASH).Methods:Methionine choline deficiency(MCD)diet combined with chronic restraint stress stimulation was used to establish the NASH rat model.After successful modeling,rats with NASH were randomly divided into model(M)group,high dose of Shugan Liqi(SG)group and low dose of Shugan Liqi(SD)group.Non-model rats of same batch were selected as normal control(N)group,and no drug intervention was allowed.In the experimental observation stage,SG group and SD group were given corresponding drugs,while M group and N group were given the same amount of distilled water.After the experiment,16S rRNA was used to detect the diversity of intestinal flora.Results:Intestinal flora OUTs value was detected.The results showed that there was no change of dominant flora at phylum level in N group,but the proportion of some flora at phylum level changed between M group and SG group.Totally 18 species belong to SG group and 7 species belong to M group.Beta diversity test showed that Bray Crutis and unweighted UniFrac were far away from each other,and there was no intersection between M group and SG group,indicating that there was a great difference in species richness between the two groups.Lefse(LDA effect SiSe)and thermogram analysis showed that there were significant differences in the abundance of intestinal flora between M group and SG group.Conclusion:Shugan Lipi Decoction can improve the species richness of intestinal flora and the proportion of species in rats with nonalcoholic steatohepatitis of liver depression and spleen deficiency syndrome.It can adjust the composition ratio of intestinal flora,affect the level of intestinal flora and restore the balance of intestinal micro ecological environment,by reducing prevotellaceae and spirillaceae and increasing clostridaceae and bacteroideae.
