摘  要:目的:探讨桔梗总皂苷对反流性食管炎(RE)大鼠食管黏膜损伤的修复作用及其机制。方法:75只SPF级SD大鼠随机分为正常对照组、模型组、西药组(兰索拉唑+莫沙必利)、桔梗总皂苷高剂量组和桔梗总皂苷低剂量组,每组15只。除正常对照组外,其余各组大鼠均建立RE模型。连续给药14 d后,采用苏木精-伊红(HE)染色观察食管黏膜病理组织学变化并进行损伤评分,ELISA法检测食管黏膜组织TNF-α、IL-6、MDA含量和SOD活力,Western blotting法和RT-PCR法检测TLR4/NF-κB信号通路蛋白及mRNA相对表达量。结果:与正常对照组比较,模型组大鼠体质量降低,死亡率增加,食管黏膜组织损伤明显,病理学评分升高,黏膜组织SOD活力降低,TNF-α、IL-6、MDA含量增加,TLR4、MyD88和NF-κB p65蛋白及mRNA相对表达量均显著增加,差异均有统计学意义(P<0.05)。与模型组比较,桔梗总皂苷高、低剂量组大鼠体质量增加,死亡率降低,食管黏膜组织损伤减轻,病理学评分降低,黏膜组织SOD活力升高,TNF-α、IL-6、MDA含量降低,TLR4、MyD88和NF-κB p65蛋白及mRNA相对表达量均减少,差异均有统计学意义(P<0.05)。结论:桔梗总皂苷能够改善反流性食管炎大鼠食管黏膜损伤,其作用机制可能与抑制TLR4/NF-κB通路有关。Objective:To explore the repair effect and mechanism of platycodon grandiflorum saponins on esophageal mucosa injury in rats with reflux esophagitis(RE).Methods:Totally 75 SPF SD rats were randomly divided into normal control group,model group,western medicine group(lansoprazole and mosapride),high-dose group of of platycodon grandiflorum saponins and low-dose group of platycodon grandiflorum saponins,with 15 rats in each group.Except for normal control group,RE models were established in all other groups.After 14 days of continuous administration,hematoxylin-eosin(HE)staining was used to observe the histopathological changes of the esophageal mucosa and score the injury.The contents of tumor necrosis factor-α(TNF-α),interleukin-6(IL-6),malondialdehyde(MDA)and superoxide dismutase(SOD)activity in esophageal mucosa were detected by enzyme linked immunosorbent assay(ELISA).The protein expression and mRNA expression of toll-like receptor 4(TLR4)/nuclear factorκB(NF-κB)signaling pathway were detected by western blotting and RT-PCR.Results:Compared with normal control group,the weight of rats decreased and the mortality increased in model group.And the model group showed more obvious esophageal mucosal tissue injury,higher pathological score,lower SOD activity,higher content of TNF-α,IL-6 and MDA,as well as higher protein expression and mRNA expression of TLR4,MyD88 and NF-κB p65 than normal control group.There were statistical differences(P<0.05).Compared with model group,the weight of rats increased and the mortality decreased in high-dose group of platycodon grandiflorum saponins and low-dose group of platycodon grandiflorum saponins.The esophageal mucosal tissue was reduced and the pathological score decreased in high-dose group of platycodon grandiflorum saponins and low-dose group of platycodon grandiflorum saponins.And the high-dose group of platycodon grandiflorum saponins and low-dose group of platycodon grandiflorum saponins showed higher SOD activity,lower content of TNF-α,IL-6 and MDA,as well as low
