摘 要:目的:探讨紫苏叶多糖对慢性阻塞性肺疾病(COPD)大鼠气道炎症及气道重塑的作用,阐明其可能作用机制。方法:采用烟熏联合脂多糖(LPS)法建立COPD大鼠模型,随机分为模型组、氯化锂(LiCl)组、紫苏叶多糖组、紫苏叶多糖+LiCl组,另取10只大鼠作为假手术组。相应干预4周后,检测用力肺活量(FVC),1秒用力呼气容积(FEV1),呼气峰流速(PEF);ELISA法检测肺泡灌洗液(BALF)中白介素-8(IL-8)、肿瘤坏死因子-α(TNF-α)水平;HE染色观察肺组织病理学变化,并计算平均肺泡数(MAN)、肺泡平均间隔内衬(MLI),测量支气管壁厚度、平滑肌厚度;Western blotting法检测肺组织无翅型MMTV整合位点家族(Wnt)5a、ras同源蛋白A(RhoA)、c-Jun氨基末端激酶(JNK)、p-JNK蛋白表达。结果:紫苏叶多糖可提高COPD大鼠肺功能,降低炎症因子水平,改善肺组织病理学变化,减少MLI,增加MAN,减少支气管壁厚度、平滑肌厚度,降低Wnt5a、RhoA、p-JNK蛋白相对表达量。在使用紫苏叶多糖基础上使用LiCl可减弱紫苏叶多糖改善效果。结论:紫苏叶多糖可抑制COPD大鼠炎症反应,减轻气道重塑,可能与抑制Wnt/PCP通路有关。Objective:To investigate the effects of perilla leaf polysaccharide on airway inflammation and airway remodeling in rats with chronic obstructive pulmonary disease(COPD),and to clarify its possible mechanism.Methods:The COPD rat model was established by the method of smoking combined with lipopolysaccharide(LPS),and the model rats were randomly divided into model group,lithium chloride(LiCl)group,perilla leaf polysaccharide group and perilla leaf polysaccharide+LiCl group.And another 10 normal rats were selected as sham operation group.After the corresponding intervention for four weeks,the forced vital capacity(FVC),forced expiratory volume in one second(FEV1)and peak expiratory flow(PEF)were measured,and the interleukin-8(IL-8)and tumor necrosis factor-α(TNF-α)level in bronchoalveolar lavage fluid(BALF)were detected by ELISA.HE staining was used to observe the pathological changes of lung tissue,and the mean alveolar number(MAN)and the mean linear intercept(MLI)were calculated.The bronchial wall thickness and smooth muscle thickness were measured.Western blotting was used to detect the expression of wingless-type MMTV integration site family(Wnt)5a,ras homologous protein A(RhoA),c-Jun N-terminal kinases(JNK)and p-JNK protein in lung tissue.Results:Perilla leaf polysaccharide can improve lung function in COPD rats,reduce the level of inflammatory factors,improve lung histopathological changes,reduce MLI,increase MAN,reduce bronchial wall thickness and smooth muscle thickness,and decrease Wnt5a,RhoA and p-JNK protein expression.On the basis of perilla leaf polysaccharide,LiCl could weaken the improvement effect of perilla leaf polysaccharide.Conclusion:Perilla leaf polysaccharide can inhibit the inflammatory response in COPD rats and reduce airway remodeling,which may be related to the inhibition of Wnt/PCP pathway.
