摘 要:目的:基于网络药理学探讨鸡血藤治疗骨关节炎的潜在作用机制。方法:通过中药系统药理学数据库(traditional Chinese medicine systems pharmacology database,TCMSP),结合口服利用度及类药性筛选药物活性成分并预测作用靶点。使用Drugbank数据库、TTD数据库、Genecard数据库、OMIM数据库获取骨关节炎相关靶点,运用韦恩图(Venny 2.1.0)取交集基因,即鸡血藤治疗骨关节炎的靶点基因。借助Cytosacape 3.7.0软件绘制"药物-活性成分-靶点"图。基于String数据库构建蛋白质相互作用网络图并通过Cytoscape 3.7.0进行分析。利用Metascape数据库对作用靶点进行GO和KEGG通路分析,并于Omicshare平台绘制气泡图。结果:共筛选出23个药物活性成分,涉及135个作用靶点;通过疾病数据库获得OA相关靶点835个;韦恩图映射出交集基因53个,富集分析得到156条KEGG通路,805条基因功能。鸡血藤的活性成分主要有β-谷甾醇、木樨草素、豆甾醇等,核心靶点包括STAT3、AKT1、TNF、VEGFA、IL-6、PTGS2、IL-1B等,主要KEGG通路有PI3K/Akt信号通路、NF-κB信号通路、IL-17信号通路等。结论:鸡血藤对骨关节炎的治疗通过多成分、多靶点、多途径实现,为今后的实验研究与药物研发奠定了一定的理论基础。Objective:To explore the potential mechanism of Jixueteng(Caulis Spatholobi) in treatment of osteoarthritis based on Internet pharmacology.Methods:To screen the active components and predict the target of action by combining oral availability and drug-like activity with traditional Chinese medicine systems pharmacology database(TCMSP).Using Drugbank database,TTD database,Genecard database,OMIM database to obtain osteoarthritis-related targets,and used Venny 2.1.0 to extract the intersection genes,which are the target genes of Caulis Spatholobi in the treatment of osteoarthritis.The graph of "drug-active component-target" was drawn with the help of Cytosacape 3.7.0 software.A protein-protein interaction network was constructed based on the String database and analyzed by Cytoscape 3.7.0.The GO and KEGG pathways were analyzed by using the Metascape database,and a bubble diagram was drawn on the Omicshare platform.Results:A total of 23 active compounds were screened,135 functional targets were involved,835 functional targets related to OA were obtained from the disease database,53 intersection genes were mapped by Wayne,156 KEGG pathways,and 805 gene functions were obtained by enrichment analysis.The main active components of Caulis Spatholobi are-sitosterol,Luteolin,stigmasterol,etc..The core targets include STAT3,AKT1,TNF,VEGFA,IL-6,PTGS2,IL-1 B,and so on,the main KEGG pathways are PI3 K/AKT,NF-κB,IL-17 and so on.Conclusion:The treatment of Caulis Spatholobi on osteoarthritis is multi-component,multi-target,and multi-approach,which lays a theoretical foundation for future experimental research and drug development.
