摘  要:甲氨蝶呤(MTX)作为一种慢作用的抗风湿药,在临床上已普遍用于治疗类风湿性关节炎。MTX在体内主要是通过其活性代谢产物多聚谷氨酸甲氨蝶呤(MTXPGs)持续发挥药理效应,其与基因多态性的关系有少量报道,并且结果并不一致。因此本文就甲氨蝶呤相关基因多态性对红细胞内MTXPGs浓度影响作一综述,探索MTX疗效与红细胞内多聚谷氨酸甲氨蝶呤浓度和基因多态性的关系,以期为临床研究提供参考。Methotrexate(MTX),as a slow-acting anti-rheumatic drug,has been widely used clinically in the treatment of rheumatoid arthritis.It is mainly in the body through its active metabolite methotrexate polyglutamates(MTXPGs)continue to exert pharmacological effects.There are few reports on its relationship with gene polymorphism,and the results are not consistent.Therefore,this article reviewed the correlation between the concentrations of erythrocyte MTXPGs and methotrexate-related gene polymorphism,explores the relationship between MTX efficacy and the concentrations of erythrocyte MTXPGs and gene polymorphism,with a view to provide reference for clinical research.
