摘  要:目的:探讨外源性硫化氢(H_(2)S)对过氧化氢(H_(2)O_(2))损伤的人脐静脉内皮细胞(HUVECs)的作用及一氧化氮(NO)系统的影响。方法:建立H_(2)O_(2)损伤HUVECs模型,观察H_(2)S的保护作用及对NO系统的影响。结果:HUVECs经H_(2)O_(2)处理后,贴壁细胞数目明显减少,细胞活力减低,内皮型一氧化氮合酶(eNOS)蛋白表达明显减弱,NO浓度明显减低;硫氢化钠(NaSH,H_(2)S供体)处理组贴壁细胞数目及细胞活力较H_(2)O_(2)模型组增加,eNOS蛋白表达较H_(2)O_(2)模型组明显增强,NO浓度较H_(2)O_(2)模型组明显提高。结论:H_(2)S通过调节NO系统部分改善H_(2)O_(2)诱导的HUVECs损伤。Objective:To explore the effect of exogenous hydrogen sulphide(H_(2)S)on treating human umbilical vein endothelial cells(HUVECs)injury induced by hydrogen peroxide(H_(2)O_(2))and the nitric oxide(NO)system in this procedure.Methods:Model of HUVECs injured by H_(2)O_(2)was established.The protective effect of H2S on the injured HUVECs was observed,and the expression of en-dothelial nitric oxide synthase(eNOS)and NO was detected.Results:The H_(2)O_(2)-treated HUVECs showed decrease adherent cell num-ber and NO concentration in cultured supernatant,reduced cell viability and eNOS protein.Compared with H_(2)O_(2)group,more in-creased number of adherent cells,elevated NO concentration in culture supernatant,increased cell viability and eNOS protein were observed in sodium hydrosulfide(NaHS,H_(2)S donor)group.Conclusion:Exogenous H_(2)S prevents HUVECs against H_(2)O_(2)-induced in-jury partially via the modulation of NO system.
