摘 要:为探究羊源多杀性巴氏杆菌(Pasteurella multocida,Pm)的生物学特性,对3份送检病死羊的肺脏组织进行细菌分离,将分离到的3株菌株分别命名为P1、P2和P3。3株菌株经多杀性巴氏杆菌特异性基因kmt PCR扩增阳性,随后检测荚膜血清型、多位点序列分型、18种毒力基因、药物敏感性和致病性。结果表明:3株分离株均为荚膜血清型D型多杀性巴氏杆菌,P1和P2序列型均为ST131, P3序列型为ST320;3株菌株的毒力基因各不相同,P1—P3均携带毒力基因RpoB、OmpH、ToxA、NanH, P3携带HgbA基因,P2携带pfhA基因,P1和P2携带SodA基因。该菌攻毒小鼠6 h后死亡,然而,采取相同的剂量攻毒白羽鸡未发生死亡;药敏结果显示3株分离株对左氧氟沙星、诺氟沙星、丁胺卡那、美洛西林、头孢曲松钠、复方新诺明敏感,对多黏菌素B、多西环素耐药。综合而言,从临床分离的3株羊源多杀性巴氏杆菌,荚膜血清型均为D型,基因型、毒力基因和致病性具有独特性,对小鼠致病力较强,对鸡致病力较弱。In order to explore the biological characteristics of Pasteurella multocida from goat, the lung tissues of three infected goats were separated for examination at different times, and the three isolates were named P1—P3. Three strains were identified as Pasteurella multocida by PCR due to kmt gene, and followed by PCR identification of capsular serotypes, Multi-site sequence typing, eighteen virulence genes, drug sensitivity and pathogenicity. The results showed that the three isolated strains belonged to the capsular serotype D of Pasteurella multocida, P1 and P2 sequence type were ST131, P3 sequence type was ST320;The three strains possessed different virulence genes, P1—P3 respectively, all isolates carried the virulence genes RpoB, OmpH, ToxA, NanH, P3 carried the HgbA gene, P2 carried the pfhA gene, P1 and P2 carried the SodA gene. The mice died after six hours of pathogenicity test with the strain, however, and no death occurred in white feather broiler chickens under the same dose. Drug susceptibility results showed that the three isolates were sensitive to levofloxacin, norfloxacin, amikacin, mezlocillin, ceftriaxone sodium, co-trimoxazole, and resistant to polymyxin B and doxycycline. In summary, the three strains of Pasteurella multocida isolated from goat, capsular serotypes are all D type. Genotype, virulence gene and pathogenicity are unique, it is higher pathogenic to mice and weaker to chickens.
