摘  要:目的:探讨小分子多肽(FNS007)对胶原诱导的关节炎大鼠T细胞亚群分型的影响。方法:采用牛CⅡ建立大鼠CIA模型,将建模成功的大鼠随机分为模型组和FNS007干预组,另选8只空白大鼠作为对照组,FNS007干预组每2 d静脉给药1次,其他两组给予等体积的PBS。给药期间观察各组大鼠状态,记录体重,对关节炎症进行评分;给药结束后处死大鼠,流式细胞仪检测外周血Th1、Th2、Treg、Th17各细胞亚群比例变化;免疫组化检测IL-17在滑膜组织的表达。结果:FNS007可改善关节炎大鼠的精神状态,与模型组比较,体重在第5、7、9天明显升高(P<0.05);FNS007可以降低关节炎大鼠的炎症评分,缓解关节炎症,与模型组比较,FNS007组大鼠炎症评分明显下降(P<0.05、P<0.01);流式结果显示,与模型组比较,FNS007组大鼠外周血Th1、Th17细胞所占比例减少(P<0.05、P<0.01),Th2、Treg细胞所占比例升高(P<0.01);免疫组化结果显示,滑膜组织中炎性细胞因子IL-17的表达下降。结论:FNS007可调节T细胞亚群的分化,降低滑膜组织IL-17表达水平,从而减轻胶原诱导性关节炎大鼠的炎症反应。Objective:To investigate the effect of small molecular polypeptide(FNS007)on the shift of T helper cells in CIA rats,and to analyze the immunological mechanism of FNS007 in the treatment of rheumatoid arthritis.Methods:Bovine typeⅡcollagen was used to establish rat rheumatoid arthritis models.Rats with swollen ankles were randomly divided into two groups:Model group and FNS007 group,and normal rats without any treatment were used as control group,with at least 8 rats in each group.Rat in therapy group were given FNS007,the normal control and model group were administrated with PBS.The state of rats in each group was observed,the weight of the body was recorded,the inflammation of joint was evaluated,and the therapeutic effect on rheumatoid arthritis rat was analyzed.All rats were killed 22 days later.T-helper type(Th1),T-helper type2(Th2),Tregulatory(Treg),T-helper type 17(Th17)in serum were detected by flow cytometry,and the expression of IL-17 in the synovial tissue was detected by immunohistochemistry.Results:FNS007 could reduce the inflammation score of arthritis rats and relieve joint inflammation.The inflammation score of rats in FNS007 group was significantly reduced compared to the model group(P<0.05,P<0.01);FNS007 could improve the living state of arthritis rats,and increase the weight of body on days 5,7,9(P<0.05);the proportion of Th1 and Th17 cells in CIA group were significantly higher than those in control group(P<0.05,P<0.01)and the proportion of Th2 and Treg cells were significantly lower than those in control group(P<0.01).Immunohistochemistry result revealed that the expression of IL-17 was decreased in FNS007 group,there were a significantly changes in synovial hyperplasia and inflammation.Conclusion:FNS007 has a significantly effect on CIA rats and the inhibition of arthritis by FNS007 may be related to maintain the balance of Th subtypes.
