摘  要:背景:间充质干细胞是具有自我更新能力和多重分化潜能的多能干细胞,能够分化为成骨细胞、脂肪细胞、软骨细胞、肌细胞和神经元等。间充质干细胞可以分泌与细胞增殖、细胞活性、免疫调节和募集有关的因子。基于这些潜能和特性,间充质干细胞可为再生医学中的细胞疗法提供理想的细胞来源。由于健康骨髓中的氧体积分数仅在3%-8%的范围内,远低于正常氧体积分数(21%),并且在受伤组织微环境中氧体积分数会更低,因此在缺氧环境中间充质干细胞的生理活性备受关注。目的:阐述缺氧状态对间充质干细胞特性以及间充质干细胞移植治疗疗效的影响。方法:检索知网、万方、PubMed等数据库2006至2020年期间关于缺氧对间充质干细胞影响的相关文献,检索词为“间充质干细胞,缺氧培养,干细胞治疗”“mesenchymal stem cells,hypoxia,regenerative therapy”等。将所查文献进行整理,排除陈旧概念和重复观点,最终选取73篇文献进行分析。结果与结论:缺氧培养可以促进间充质干细胞增殖、提高迁移能力、调控细胞分化。在临床治疗中通过对间充质干细胞进行缺氧预处理可调节其分化能力,提高细胞存活率,加速组织愈合,改善治疗效果。缺氧培养对于间充质干细胞的调控作用为骨质疏松临床治疗和移植再生治疗探究提供了新思路。BACKGROUND:Mesenchymal stem cells are multipotent stem cells owing the ability of self-renewal and multiple differentiation potential,which promise mesenchymal stem cells to differentiate into several cell types,including osteoblasts,adipocytes,chondrocytes,myocytes and neurons and so on.Mesenchymal stem cells can secrete factors associated with cell proliferation and survival,immunomodulation,and recruitment.These potentials and characteristics make mesenchymal stem cells an ideal cell source for cell-based treatment of tissues repair in regenerative medicine.Since the oxygen constitution only at range of 3%-8%in healthy bone marrow which is much lower than the normal oxygen concentration of 21%,and this concentration will become lower in the injured tissue microenvironment.Thus,the fate of mesenchymal stem cells in hypoxic environment attracts lots of attention.OBJECTIVE:To explicate the roles of hypoxic condition in mesenchymal stem cells properties and the therapeutic effect of mesenchymal stem cells transplantation.METHODS:The articles about effect of hypoxia on mesenchymal stem cell were searched in CNKI,Wanfang,and PubMed from 2006 to 2020.The search key words were“mesenchymal stem cells,hypoxia culture,stem cell therapy”in Chinese,and“mesenchymal stem cells,hypoxia,regenerative therapy”in English.The collected literature was sorted out to eliminate obsolete concepts and repeated views,and 73 articles were finally selected for analysis.RESULTS AND CONCLUSION:Hypoxic culture could promote cell proliferation,migration ability of mesenchymal stem cells and regulate cell differentiation.In clinical treatment,hypoxic preconditioning could regulate the differentiation ability of mesenchymal stem cells,improve cell survival rate,accelerate tissue healing,and improve the therapeutic effect.Hypoxic culture on the regulation of mesenchymal stem cells provides a new idea for the clinical treatment of osteoporosis and transplantation regenerative therapy.
