摘 要:旨在探究组蛋白细胞周期调节(Histone cell cycle regulator,HIRA)基因g.71874104G>A和g.71833755T>C位点对绵羊排卵数的影响,为绵羊高排卵数主效基因的筛选提供基因来源。筛选出含g.71874104G>A和g.71833755T>C两个突变位点的不同基因型小尾寒羊母羊,并在同期发情之后利用活体腹腔镜观测法对排卵数进行测定。同时,利用放射免疫法对绵羊血清中促卵泡素(Follicle-stimulating hormone,FSH)、促黄体素(Luteinizing hormone,LH)、孕酮(Progesterone,P4)以及雌二醇(Estradiol,E2)浓度进行测定,并分析2个位点突变对绵羊血清激素分泌的影响。结果表明,HIRA基因的2个突变位点对绵羊个体排卵数和血清中生殖激素FSH、LH和P4的分泌均无显著影响(P>0.05),但对E2的分泌有显著影响(P<0.05)。综上可得,HIRA基因的g.71874104G>A和g.71833755T>C两个突变位点可能不是影响小尾寒羊排卵数的关键位点。To explore the effect of g.71874104G>A and g.71833755T>C sites of histone cell cycle regulator(HIRA)gene on ovu-lation number in sheep,Small Tail Han ewes containing two mutation sites g.71874104G>A and g.71833755T>C were genotyped and selected in the study,then the ovulation rate was detec-ted and recorded using laparoscopic observation on living sheep after estrus synchronization.At the same time,the concentrations of follicle stimulating hormone(FSH),luteinizing hormone(LH),progesterone(P4)and estradiol(E2)in sheep serum were detected by radioimmunoassay,and effects of two mutations on hormone secretion in sheep were analyzed.The results showed that the two mutations in HIRA gene had no significant effect on the ovulation rate and the secretion of FSH,LH and P4 in serum,but both had a significant effect on the secretion of E2.In summary,the g.71874104G>A and g.71833755T>C mutation sites of HIRA gene may not be the key loci that affect the ovulation number of Small Tail Han sheep.
