大同黄花菜茎段组织培养研究

摘　　要：以大同黄花菜种子培养形成的无菌苗的根、茎、叶为外植体进行组织培养。结果表明,种子去皮后灭菌和萌发效果最好,二次灭菌可消除萌发期间的污染,萌发率达100%。无菌苗的根、茎、叶切段接种培养后,仅茎段诱导形成了不定芽,其中MS培养基的诱导率最高,达87.5%,继代培养全部顺利抽生叶片,生根培养有45%发生根系,形成了完整的试管苗。根和叶的切段在诱导培养基上没有任何形态变化。说明黄花菜无菌苗的茎段作为外植体进行组织培养是可行的,根和叶的切段难以诱导形成组培苗。In this study,the roots,stems and leaves of the sterile seedlings of daylily in Datong were used as explants for tissue culture.The results showed that the sterilizing and germinating effect was the best after removing seed coat,and the contamination during the germination period could be eliminated by secondary sterilization with 100%germinating rate.After inoculation and culture of roots,stems and leaves,only adventitious buds were induced from stem segments,and the induction rate of MS medium was the highest,reaching 87.5%.And,the segments grew leaves successfully,and 45%of adventitious buds generated root system,forming a complete test-tube seedling during subculture.Also,root and leaf segments did not show any morphological changes on induction medium,which indicated that it was feasible to use stem segments as explants for tissue culture.In addition,it was difficult to induce tissue culture seedlings from root and leaf segments,which provided experimental basis for the establishment of tissue culture system of daylily.
