小白菊内酯及联合5-氟尿嘧啶注射液对鼠结肠癌CT-26细胞凋亡和增殖的作用研究

摘　　要：目的:探讨小白菊内酯联合5-氟尿嘧啶注射液(5-FU)对鼠结肠癌CT-26细胞增殖及凋亡的影响。方法:不同浓度的小白菊内酯和5-FU分别处理CT-26细胞,CCK-8法以及克隆形成检测细胞增殖的变化情况,流式细胞学以及蛋白质印迹法(Western blot)检测细胞凋亡情况。结果:小白菊内酯和5-FU对CT-26细胞均具有抑制作用且呈浓度依赖性,5μmol/L 5-FU联合10μmol/L小白菊内酯作用时抑制CT-26细胞增殖并诱导细胞凋亡。结论:小白菊内酯联合5-FU抑制CT-26细胞增殖并诱导细胞凋亡。Objective:To investigate the effect of parthenolide and(or) 5-FU on proliferation and apoptosis in human colon cancer cell CT-26.Methods:After treated by parthenolide and(or) 5-FU with different concentrations,the proliferation of CT-26 cells was determined by CCK-8 assay and Clone formation,and the cell apoptosis was examined by flow cytometry and western blot.Results:Parthenolide and 5-FU were capable of inhibiting CT-26 cells proliferation,and the inhibition was dose dependent.Parthenolide5 μmol/L and 5-FU 5 μmol/L synergistically inhibited the proliferation of CT-26 cells.The cell apoptosis also could be induced by parthenolide and(or) 5-FU.Conclusion:Parthenolide and 5-FU have synergistical effects on inhibiting cell proliferation and inducing cell apoptosis of CT-26 cells.
