基于UPLC-Q-TOF-MS技术的清热消炎宁胶囊化学成分分析

摘　　要：目的采用超高效液相色谱-四级杆-飞行时间质谱(UPLC-Q-TOF-MS)对清热消炎宁胶囊中的化学成分进行分析和鉴定。方法采用Waters Acquity HSS T3色谱柱(50 mm×2.1 mm,1.8μm),以乙腈-水(各含0.1%甲酸)为流动相进行梯度洗脱,流速0.5 mL·min^(-1),柱温35℃,选择电喷雾离子源(ESI)、负离子模式下检测,通过与对照品比对和质谱数据分析及结合文献数据的方法分析化学成分。结果初步在清热消炎宁胶囊中检识到42个化合物,包括13个咖啡酰类衍生物,7个香豆素类成分,5个黄酮类成分,12个倍半萜类化合物及5个有机酸类成分。结论UPLC-Q-TOF-MS联用技术为鉴别清热消炎宁胶囊中化学成分提供简便、快速、准确的方法,为进一步分析清热消炎宁胶囊的物质基础及质量控制提供了新技术方法。OBJECTIVE To analyze and identify the chemical compositions in Qingrexiaoyanning capsule by UPLC-Q-TOF-MS.METHODS The separation was performed on a Waters Acquity UPLC HSS T3 column(50 mm×2.1 mm,1.8μm)by gradient elution with acetonitrile-water(both containing 0.1%formic acid)as mobile phase at a flow rate of 0.5 mL·min^(-1).The column temperature was set at 35℃.The ion was achieved using ESI in negative mode.The chemical compositions were analyzed by comparison with reference substances and mass spectrometry data analysis and literature data.RESULTS A total of 42 compositions in Qingrexiaoyanning capsule were identified,including thirteen caffeoyl derivatives,seven coumarins,five flavonoids,twelve terpenoids and five organic acids.CONCLUSION This study shows that UPLC-Q-TOF-MS technology provides a simple,rapid,and accurtate method for the identification of chemical constituents in Qingrexiaoyanning capsule,which provides a new technology method for the pharmacological basis and quality control of Qingrexiaoyanning capsule.
