摘  要:目的探讨动脉粥样硬化小鼠主动脉组织中CXC型趋化因子配体16(CXCL16)和MMP-1的表达。方法选取8周龄的SPF级ApoE基因敲除(ApoE-/-)雄性小鼠10只,给予高脂饲料喂养构建动脉粥样硬化模型(实验组),另选取8周龄的SPF级雄性小鼠10只给予普通饲料喂养作为对照组。两组小鼠均在喂养8周后处死,将心脏-主动脉整条分离。采用Western blot法检测两组小鼠主动脉组织中CXCL16和MMP-1蛋白表达。采用RT-PCR法检测主动脉组织中CXCL16和MMP-1 mRNA表达。结果实验组小鼠主动脉组织CXCL16和MMP-1蛋白表达量高于对照组(P<0.05)。实验组小鼠主动脉组织CXCL16和MMP-1 mRNA表达量高于对照组(P<0.05)。结论动脉粥样硬化小鼠主动脉组织中高表达炎症因子CXCL16和MMP-1。Objective To explore the expressions of CXC-chemokine ligand 16(CXCL16)and matrix metalloproteinase-1(MMP-1)in the aortic tissues of the mice with atherosclerosis.MethodsThe atherosclerosis model(group A)was established by high fat feeding in 10 male mice aged 8 weeks old with apolipoprotein E knockout(ApoE-/-)and another 10 SPF male mice aged 8 weeks old(group B)were fed with ordinary feed as the controls.Both groups of mice were sacrificed 8 weeks later and the whole heart and aorta were separated.Western blot and RT-PCR were used to detect the protein and mRNA expressions of CXCL16 and MMP-1 in the aorta tissues.Results The protein expressions of CXCL16 and MMP-1 in the aorta tissues in group A were higher than those in group B(P<0.05).The mRNA expressions of CXCL16 and MMP-1 in the aorta tissues in group A were higher than those in group B(P<0.05).Conclusion The inflammatory factors CXCL16 and MMP-1 are highly expressed in the aortic tissues of the mice with atherosclerosis.
