摘 要:[目的]探讨发酵人参对H4IIE大鼠肝癌细胞三酰甘油蓄积的抑制作用机制.[方法]用0、25、50、100、200 mg/L的发酵人参处理H4IIE大鼠肝癌细胞24 h,采用MTS法测定发酵人参对H4IIE细胞生存率的影响,并测定三酰甘油(TG)水平,采用Western blot法检测发酵人参对AMPK、ACC、p-AMPK及p-ACC蛋白表达的影响.[结果]发酵人参在0~200 mg/L质量浓度范围内对H4IIE大鼠肝癌细胞生长率未见产生影响,在0~100 mg/L质量浓度范围内剂量依赖性地降低细胞中的三酰甘油水平(P<0.001).发酵人参显著增加AMP依赖的蛋白激酶及其下游激酶ACC的磷酸化.[结论]发酵人参可能通过AMPK信号通路调控H4IIE大鼠肝癌细胞三酰甘油的蓄积.OBJECTIVE To explore the inhibitory mechanism of fermented ginseng on triglyceride accumulation in H4 IIE rat hepatoma cell.METHODS H4 IIE rat hepatoma cells were treated with 0,25,50,100,200 mg/L fermented ginseng for 24 hours.MTS assay was used to determine the effect of fermented ginseng on the survival rate of H4 IIE cells,and the level of triglyceride(TG)were measured.Western blot method was used to detect the effects of fermented ginseng on AMPK,ACC,p-AMPK and p-ACC protein expression.RESULTS Fermented ginseng had no effect on the growth rate of H4 IIE rat hepatoma cell in the range of 0-200 mg/L,and in the range of 0-100 mg/L,the level of TG in cells was dose-dependently decreased(P<0.001).Fermented ginseng significantly increased the phosphorylation of AMPK and its downstream kinase ACC.CONCLUSION Fermented ginseng may regulate the accumulation of TG in H4 IIE rat hepatoma cells through AMPK signaling pathway.
