摘  要:抽穗期是小麦生产中重点关注的农艺性状,与生殖器官发育密切相关,而GRF基因是植物器官建成的调控因子,为了明确小麦GRF基因调控抽穗期的生物学功能,本研究利用生物信息学方法分析小麦TaGRF1基因与水稻OsGRF1基因的亲缘关系,利用VIGS病毒沉默技术鉴定TaGRF1生物学功能,借助RT-qPCR对沉默株系中抽穗相关基因VRN1、VRN2、SOC1进行表达分析。结果表明,TaGRF1与水稻OsGRF1基因同源性为53.1%~71.56%,TaGRF1沉默后小麦抽穗显著延迟,沉默株系中的TaGRF1基因与对照相比显著下调表达,小麦调控抽穗期关键基因VRN1也显著下调表达,同时,VRN2基因上调表达,而SOC1基因表达无显著变化。本研究结果表明,小麦TaGRF1基因可能通过调控抽穗期关键基因VRN1、VRN2的表达促进小麦抽穗。Headin g date is a key agronomic trait concerned in wheat production.It is closely related to the development of reproductive organs.GRF gene is a regulatory factor in organ morphogenesis process.In order to clarify the biological function of wheat GRF gene in regulating heading date,the genetic relationship between wheat TaGRF1 gene and rice OsGRF1 gene was analyzed by bioinformatics method.The biological function of TaGRF1 was studied using VIGS virus silencing technology.The expression pattern of the flowering-related genes VRN1,VRN2,SOC1 was determined using RT-qPCR.The results showed that the similarity between TaGRF1 and OsGRF1 genes was 53.1%~71.56%.The heading date was significantly delayed after TaGRF1 silencing.The expression of TaGRF1 gene in the silenced lines was significantly down-regulated compared with the control.The expression of VRN1 which regulates heading period was also significantly down regulated.Another heading dated related gene VRN2 gene was up-regulated,while the expression of SOC1 gene remained unchanged.The results of this study show that wheat Ta GRF1 gene may promote wheat heading by regulating the expression of heading data-related genes VRN1 and VRN2.
