摘 要:棉蚜是继棉铃虫之后棉花的又一主要害虫。为了验证转多凝集素基因的陆地棉对蚜虫的抗性,本研究选取了研究广泛且具有良好抗蚜虫效果的雪花莲凝集素和苋菜凝集素构建双抗虫基因的植物表达载体,通过农杆菌介导法转入棉花R15中,共获得6株T_(0)代转基因植株。分别通过PCR、RT-qPCR和Western blot证实了双凝集素基因在棉花中的遗传和表达。通过在转基因植株上接种棉蚜的生物测定,发现转凝集素基因的棉花对棉蚜具有一定的抑制作用,抑制率可达36%~55.5%,抗虫效果明显优于对照植株。本实验采用环境友好型、安全性较高的凝集素基因,既可以减少蚜虫的危害,降低棉花的生产成本,提升棉花的质量和产量,还可以保护非靶标昆虫,减少化学农药的使用,保护生态环境。Cotton aphid is another major pest of cotton after cotton bollworm.In order to test the resistance of upland cotton with polylectin gene to aphids.In this study,Galanthus nivals agglutinin and Amaranthus caudatus agglutinin,which have been widely studied and have good resistance to aphids,were selected to construct plant expression vectors of double resistance genes.They were transferred into cotton R15 by agrobacterium-mediated method,and a total of 6 transgenic plants of _(T)0 generation were obtained.The inheritance and expression of lectins were confirmed by PCR,RT-q PCR and Western blot analysis,respectively.An aphid bioassay on transgenic cotton revealed that transgenic cottons had an apparent inhibition compared with R15.The inhibition rate is 36%~55.5%.In this experiment,environmentally friendly and highly safe lectin genes were used,which can not only reduce the harm of aphids,reduce the cost and improve the quality and yield,but also protect non-target insects,reduce the use of chemical pesticides,and protect the ecological environment.
